決奈達隆對新生大鼠心室肌細胞HCN通道m(xù)RNA和蛋白表達的影響

【摘要】:目的通過檢測新生大鼠心室肌細胞在給予藥物決奈達隆前后超極化激活環(huán)核苷酸門控陽離子通道(HCN通道)mRNA和蛋白水平的變化,探討決奈達隆對HCN通道表達的影響。方法分離新生SD大鼠心室肌,Ⅱ型膠原酶消化,通過差速貼壁分離法收集獲得單一的心室肌細胞。并根據(jù)濃度(0.1、0.5、1.0、5.0、10.0、20.0μmol/L的決奈達隆對細胞進行處理48h)和時間(濃度為10μmol/L的決奈達隆對細胞進行1、6、12、24、48h的處理)梯度分組。采用實時熒光定量PCR法和Western blot檢測HCN2、HCN4通道m(xù)RNA水平和蛋白水平。結果濃度梯度組和時間梯度組的HCN2 mRNA和HCN4 mRNA表達水平均低于對照組(P0.05);與對照組比較,10μmol/L決奈達隆處理后12h組的蛋白水平明顯下調(P0.01)。結論決奈達隆可抑制HCN2、HCN4通道m(xù)RNA和蛋白的表達,且作用呈濃度依賴性,在給藥后12h達到最大作用。
