不同材質(zhì)濾膜對(duì)于加替沙星滴眼液中抑菌劑的吸附性研究

【摘要】:目的:考察加替沙星滴眼液除菌過(guò)濾時(shí)不同材質(zhì)濾膜對(duì)于苯扎氯銨的吸附性;建立高效液相色譜法測(cè)定加替沙星滴眼液中苯扎氯銨的含量。方法:采用材質(zhì)為聚醚砜(PES)、聚偏二氟乙烯(PVDF)的濾膜過(guò)濾加替沙星滴眼液后,收集濾液,于100,500,1 000,1 500,2 000 mL時(shí)取樣,測(cè)定過(guò)濾后苯扎氯銨的含量;以水(用1 mol·L-1高氯酸調(diào)pH至2.2±0.05)-乙腈-異丙醇(58∶33∶9)為流動(dòng)相等度洗脫;流速1.0mL·min-1;柱溫40℃;檢測(cè)波長(zhǎng)214 nm。結(jié)果:材質(zhì)為聚偏二氟乙烯(PVDF)的濾膜對(duì)于苯扎氯銨有6.3%的吸附,而材質(zhì)聚醚砜(PES)對(duì)于苯扎氯銨無(wú)吸附性;苯扎氯銨在11.1~111μg·mL-1(222~2 220ng)濃度范圍內(nèi)呈良好的線性,線性方程為y=22 162x-15 620(r=0.999 9),平均回收率為100.58%、RSD=0.60%(n=9),準(zhǔn)確度較好。結(jié)論:對(duì)于加替沙星滴眼液的生產(chǎn),采用材質(zhì)為聚醚砜(PES)的濾膜對(duì)其中抑菌劑苯扎氯銨無(wú)吸附;建立了HPLC法測(cè)定加替沙星滴眼液中苯扎氯銨的方法,操作簡(jiǎn)便,靈敏度高,重復(fù)性好,可快速準(zhǔn)確用于加替沙星滴眼液中苯扎氯銨含量分析。
