二氫楊梅素對(duì)濕性年齡相關(guān)性黃斑變性的影響
[Abstract]:Objective: to investigate the effect of dihydromyricetin (DMY) on (AMD) of wet age related macular degeneration. Methods: the AMD model was established by photocoagulation on the fundus of adult Japanese white rabbits with 532 nm laser. After 4 weeks of photocoagulation, DMY (100 mg kg~ (-1) d ~ (-1) was administered intragastrically. The levels of vascular endothelial growth factor (VEGF) and pigment epithelium-derived factor (PEDF) in peripheral blood were detected by enzyme-linked immunosorbent assay (ELISA). At the end of the experiment, hematoxylin-eosin staining was used to detect the choroidal neovascularization (CNV); in experimental animals, and the indexes of liver, kidney and spleen were calculated. Results: after 4 weeks of administration, the level of VEGF in peripheral blood of model group was 63.3%, while that of model control group was 48.6%. After 8 weeks of administration, the decrease of VEGF in peripheral blood of model group was 46.7%, and that of model control group was 20.1%. After 4 weeks of administration, the decrease of PEDF in peripheral blood of model group was 55.9, that of model control group was 58.6, and that of peripheral blood PEDF was 12.1 in model group and 53.1 in model control group after 8 weeks of administration. After 8 weeks of administration, significant CNV, was found in the model control group, but no significant CNV; was found in the model administration group. The liver index and spleen index in the model group were significantly lower than those in the model control group (P < 0. 147 and P < 0. 223), but there was no significant difference between the model group and the model control group (P < 0. 874). Conclusion: DMY has a certain pharmacological effect on inhibiting the growth of CNV in Japanese large ear white rabbits with wet AMD model, and its mechanism may be related to its down-regulation of VEGF expression, up-regulation of PEDF expression, invigorating qi and activating blood circulation, and invigorating spleen and soothing liver.
