【摘要】：探討蒿甲醚與依托泊苷聯(lián)用對(duì)小細(xì)胞肺癌細(xì)胞H446增殖及侵襲能力的抑制作用。采用MTT法檢測(cè)蒿甲醚、依托泊苷單用及聯(lián)合作用對(duì)H446細(xì)胞增殖的影響;采用Annexin V/PI熒光雙染法檢測(cè)蒿甲醚、依托泊苷單用及聯(lián)合作用對(duì)H446細(xì)胞凋亡的影響;采用PI單染法檢測(cè)蒿甲醚、依托泊苷單用及聯(lián)合作用對(duì)H446細(xì)胞周期分布的影響。采用基質(zhì)膠侵襲實(shí)驗(yàn)檢測(cè)蒿甲醚、依托泊苷單用及聯(lián)合作用對(duì)H446侵襲能力的影響。結(jié)果表明,蒿甲醚、依托泊苷單用以及聯(lián)合使用均能有效抑制H446的增殖,且呈劑量依賴性,兩藥聯(lián)用具有協(xié)同作用。蒿甲醚單用不能明顯促進(jìn)細(xì)胞凋亡,但與依托泊苷(300 nmol/L)聯(lián)用72 h可以比較明顯促進(jìn)H446凋亡,具有統(tǒng)計(jì)學(xué)意義(P0.05)。蒿甲醚單用對(duì)H446的細(xì)胞周期沒有顯著影響,聯(lián)合作用48 h對(duì)細(xì)胞周期G_2期阻滯主要是依托泊苷的作用。單用蒿甲醚、依托泊苷以及二者聯(lián)合使用都能顯著抑制H446的侵襲能力。
[Abstract]:To investigate the inhibitory effect of artemether and etoposide on the proliferation and invasion of small cell lung cancer cell line H446. The effects of artemether, etoposide alone and combined action on the proliferation of H446 cells were detected by MTT assay, and the effects of artemether and etoposide alone and combined action on the apoptosis of H446 cells were detected by Annexin V/PI fluorescence double staining. The effects of artemether, etoposide alone and combined action on cell cycle distribution of H446 cells were detected by PI single staining. The effects of artemether, etoposide alone and combined action on the invasion ability of H 446 were detected by matrix gel invasion test. The results showed that artemether etoposide alone and combined use could effectively inhibit the proliferation of H446 in a dose-dependent manner. Artemether alone could not significantly promote apoptosis, but combined with etoposide (300 nmol/L) for 72 h could significantly promote apoptosis of H446 (P0.05). Artemether alone had no significant effect on the cell cycle of H446. The combined action of 48 h on cell cycle arrest was mainly caused by etoposide. Artemether alone, etoposide and their combination could significantly inhibit the invasion ability of H 446.
