丙戊酸對百草枯與脂多糖誘導(dǎo)的巨噬細(xì)胞炎性極化的作用

【摘要】:目的分析組蛋白去乙;(HDAC)抑制劑丙戊酸(VPA)對百草枯(PQ)和脂多糖(LPS)誘導(dǎo)的巨噬細(xì)胞炎性極化的作用。方法小鼠巨噬細(xì)胞RAW264.7 37℃、5%CO2條件下培養(yǎng),穩(wěn)定傳代后分組,分別給予以下處理:(1)PQ;(2)PQ+VPA(HDACⅠ和Ⅱa類抑制劑);(3)PQ+Apicidin(HDACⅠ類抑制劑);(4)PQ+MC1568(HDACⅡa類抑制劑);(5)LPS;(6)LPS+VPA;(7)LPS+Apicidin;(8)LPS+MC1568。8 h后收集各組細(xì)胞上清及細(xì)胞團(tuán),RT-PCR、ELISA及流式細(xì)胞檢測巨噬細(xì)胞表型標(biāo)志物的表達(dá)。結(jié)果 PQ與LPS均促進(jìn)巨噬細(xì)胞向促炎表型極化;VPA、Apicidin和MC1568均抑制PQ、LPS誘導(dǎo)巨噬細(xì)胞表型極化作用,但抑制作用不完全相同。結(jié)論 VPA抑制PQ和LPS誘導(dǎo)的巨噬細(xì)胞促炎活性,但對PQ和LPS的作用方式各有特點。
