環(huán)丙沙星體外誘導(dǎo)銅綠假單胞菌對(duì)群體感應(yīng)系統(tǒng)正常與缺陷最低抑菌濃度的影響

【摘要】:目的:探究群體密度(QS)系統(tǒng)正常與缺陷的銅綠假單胞菌(PA)通過(guò)環(huán)丙沙星誘導(dǎo)后對(duì)其最低抑菌濃度(MIC)影響的區(qū)別。方法:將PA按照前期研究的基因測(cè)序方法分成正常組和缺陷組。采用瓊脂倍比稀釋法測(cè)定對(duì)環(huán)丙沙星的MIC值,用1/2MIC、1MIC、2MIC、4MIC的環(huán)丙沙星各誘導(dǎo)5代,保存1代、3代和5代的菌株。誘導(dǎo)濃度和誘導(dǎo)代數(shù)對(duì)MIC值的影響采用重復(fù)測(cè)量方差分析方法,正常組與缺陷組的比較采用秩和檢驗(yàn)。結(jié)果:兩組組內(nèi)比較:在同一濃度下,2組各隨著誘導(dǎo)代數(shù)增加MIC值均增加(P均0.05);在同一代數(shù)下隨著誘導(dǎo)濃度的增加,正常組第3代和第5代的MIC值也增加(P均0.05),缺陷組第3代和第5代的MIC值差異有統(tǒng)計(jì)學(xué)意義(P均0.05)。2組組間比較:QS系統(tǒng)分組與誘導(dǎo)代數(shù)無(wú)交互效應(yīng)(P0.05),且缺陷菌株MIC值都低于正常菌株;與誘導(dǎo)濃度存在交互效應(yīng)(P0.05)。固定代數(shù)固定濃度兩組MIC值比較:在1MIC誘導(dǎo)的第3代和第5代,2組之間有統(tǒng)計(jì)學(xué)差異(P0.05)。結(jié)論:不同誘導(dǎo)濃度對(duì)QS系統(tǒng)正常組和缺陷組MIC值的影響有區(qū)別。
[Abstract]:Aim: to investigate the difference between normal and defective Pseudomonas aeruginosa (PA) induced by ciprofloxacin (ciprofloxacin) on its minimum inhibitory concentration (MIC). Methods: PA was divided into normal group and defective group according to the previous gene sequencing method. The MIC value of ciprofloxacin was determined by Agar doubling dilution method. The ciprofloxacin of 1 / 2MIC1 MIC2MIC2MIC4MIC was induced for 5 generations each, and the strains of the third and fifth generations were preserved in the first generation of ciprofloxacin. The influence of induced concentration and induction algebra on MIC value was analyzed by repeated measurement variance, and rank sum test was used to compare normal group with defect group. Results: the MIC values of each group increased with the increase of induction algebra at the same concentration (P < 0. 05), and increased with the increase of induced concentration under the same algebra. The MIC values of the third and fifth generations of the normal group were also increased (P 0.05), and the MIC values of the third and fifth generation of the defect group were significantly different (P 0.05). 2 there was no interaction between the group of groups with the induced algebra and the group of groups (P0.05), and the MIC of the defective strain of the group was higher than that of the control group (P0.05). The values were lower than those of normal strains. There was an interaction effect with the induced concentration (P0.05). Comparison of MIC values between the two groups of fixed algebraic concentration: there was statistical difference between the third generation and the fifth generation group induced by 1MIC (P0.05). Conclusion: the effects of different inductive concentrations on the MIC values of normal and defective QS systems are different.
