頂空氣相色譜法測定苯甲酸阿格列汀原料藥中3種有機(jī)溶劑殘留量

【摘要】:目的:建立檢測苯甲酸阿格列汀原料藥中甲苯、正丁醇、乙醇?xì)埩袅康姆椒。方?采用頂空氣相色譜法。采用氫火焰離子化檢測器,色譜柱為6%氰丙基苯基-94%甲基聚硅氧烷毛細(xì)管柱,進(jìn)樣口溫度為200℃,檢測器溫度為250℃,分流比為10∶1,流速為1 ml/min,頂空進(jìn)樣,進(jìn)樣量為20μl,程序升溫;以外標(biāo)法計(jì)算甲苯、正丁醇、乙醇的殘留量。結(jié)果:3種有機(jī)溶劑在16 min內(nèi)能完全分離,分離度均大于1.5;甲苯、正丁醇、乙醇的質(zhì)量濃度分別在22.25~178、125~1 000、125~1 000μg/ml范圍內(nèi)與各自峰面積呈良好的線性關(guān)系(r=0.999 8、0.999 6、0.998 7);精密度試驗(yàn)的RSD≤1.36%;甲苯、正丁醇、乙醇的平均加樣回收率分別為98.2%、101.8%、98.90%,RSD分別為0.6%、0.7%、0.9%(n均為9)。結(jié)論:該方法操作簡便、結(jié)果準(zhǔn)確,可用于苯甲酸阿格列汀原料藥中有機(jī)溶劑殘留量的控制。
