過氧化氫通過內(nèi)質(zhì)網(wǎng)應激信號通路對Hela細胞自噬和凋亡的誘導作用

【摘要】:目的:利用過氧化氫(H2O2)構建人宮頸癌Hela細胞體外氧化應激模型,觀察其對Hela細胞自噬和凋亡的影響,并探討H2O2與內(nèi)質(zhì)網(wǎng)應激(ERS)信號通路之間的關系。方法:以1mmol·L-1 H2O2構建體外氧化應激模型;實驗分為對照組、H2O24h組、H2O28h組和H2O212h組,各組細胞分別以H2O2作用0、4、8及12h后,利用Western blotting法檢測各組Hela細胞中ERS及凋亡、自噬相關蛋白表達。結果:與對照組比較,各H2O2組Hela細胞中Cleaved Caspase-3表達水平升高(P0.05),Cleaved Caspase-4表達水平亦明顯升高(P0.05);各H2O2組Hela細胞中LC3-Ⅱ表達水平高于對照組(P0.05),GRP78表達水平亦高于對照組(P0.05);隨著H2O2作用時間的延長,各H2O2組Hela細胞中IRE1a表達水平明顯升高(P0.01),p-JNK表達水平明顯高于對照組(P0.05)。結論:在H2O2所構建的體外氧化應激模型中H2O2能夠誘導Hela細胞發(fā)生自噬和凋亡,且該誘導作用由ERS信號通路介導。
[Abstract]:Aim: to investigate the effects of hydrogen peroxide (H _ 2O _ 2) on autophagy and apoptosis of human cervical cancer Hela cells in vitro, and to explore the relationship between H2O2 and endoplasmic reticulum stress (ERS) signaling pathway. Methods: 1mmol L-1 H 2O 2 was used to construct oxidative stress model in vitro, and the cells were divided into two groups: control group (H 2O 2 24 h), H 2O 2 group (H 2O 2 28 h) and H 2O 2 group (H 2O 2 12 h). After treated with H 2O 2 for 0 h and 12 h respectively, the expression of ERS, apoptosis and autophagy related protein in Hela cells were detected by Western blotting assay.
