【摘要】：目的觀察氯吡格雷家兔血小板聚集率與創(chuàng)傷性出血的影響。方法選取21只健康家兔,隨機(jī)分為3組,A組為對照組,給予生理鹽水5 ml·kg-1·d-1灌胃,B、C組給予濃度為0.575 mg/ml的氯吡格雷藥液5 ml·kg-1·d-1灌胃,B組連續(xù)灌胃7 d,C組連續(xù)灌胃14 d。檢測3組家兔服藥前、服藥7、14 d的血小板聚集率、血小板計(jì)數(shù),并檢測各組家兔服藥14 d的出血時(shí)間、凝血時(shí)間,記錄創(chuàng)傷的出血量。結(jié)果 3組家兔服藥前血小板聚集率及血小板計(jì)數(shù)無明顯差異(P0.05)。服藥后第7天,B、C組家兔血小板聚集率較A組明顯降低(P0.001);第14天,B組家兔血小板聚集率與A組比較無顯著差異(P0.05),出血時(shí)間無明顯延長,創(chuàng)傷性出血量無明顯增加(P0.05)。第14天C組血小板聚集率與A、B組相比顯著降低(P0.001),C組的出血時(shí)間較A、B組顯著延長(P0.01),創(chuàng)傷性出血量較A、B組有所增加,但無統(tǒng)計(jì)學(xué)差異(P0.05)。B、C組家兔凝血時(shí)間及血小板計(jì)數(shù)與A組比較無顯著差異(P0.05)。創(chuàng)傷性出血量與血小板聚集率無顯著相關(guān)性。結(jié)論不停用常規(guī)劑量的氯吡格雷未明顯增加創(chuàng)傷性出血量;血小板聚集率暫不能作為術(shù)前停藥的參考指標(biāo)。
[Abstract]:Objective to observe the effect of clopidogrel on platelet aggregation and traumatic hemorrhage in rabbits. Methods Twenty-one healthy rabbits were randomly divided into three groups as control group. The control group was given normal saline 5 ml kg-1 d-1 intragastric infusion of clopidogrel solution 5 ml kg-1 d -1 for 14 days. The platelet aggregation rate, platelet count, bleeding time, coagulation time and wound bleeding volume were measured in 3 groups of rabbits before taking medicine for 7 days and 14 days after taking medicine. Results there was no significant difference in platelet aggregation rate and platelet count between the three groups (P 0.05). The platelet aggregation rate in group B was significantly lower than that in group A on day 7 (P0.001), but there was no significant difference between group B and group A on the 14th day (P0.05), the bleeding time was not significantly prolonged, and the amount of traumatic hemorrhage was not significantly increased (P0.05). On the 14th day, the platelet aggregation rate in group C was significantly lower than that in group A (P 0.001). The bleeding time in group C was significantly longer than that in group A (P 0.01), and the amount of traumatic bleeding was higher than that in group A (P 0.01). But there was no statistical difference (P0.05). There was no significant difference in coagulation time and platelet count between group A and group B (P0.05). There was no significant correlation between traumatic blood loss and platelet aggregation. Conclusion Clopidogrel does not increase the amount of traumatic blood loss and platelet aggregation rate can not be used as a reference index for withdrawal of drugs before operation.
