5-氟尿嘧啶直腸溫敏膠膜劑的制備

【摘要】:目的:制備5-氟尿嘧啶(5-FU)直腸溫敏膠膜劑。方法:以泊洛沙姆407(P407)和泊洛沙姆188(P188)為溫敏材料制備5-FU直腸溫敏膠膜劑,以溶液-凝膠的相變溫度、凝膠強(qiáng)度、生物黏附力為指標(biāo),對(duì)溫敏材料的比例、5-FU質(zhì)量濃度、p H、生物黏附劑種類進(jìn)行篩選。結(jié)果:以P407-P188(17∶2.5)為溫敏材料,2%卡波姆940為生物黏附劑,p H為7.1,制得規(guī)格為500 mg/30 ml的5-FU溫敏凝膠劑。所制凝膠在室溫下為自由流動(dòng)的液體,在生理?xiàng)l件下相變形成膠膜,相變溫度為35~36.6℃,凝膠強(qiáng)度為20~60 g,生物黏附力為20~100 g,均符合直腸應(yīng)用要求。結(jié)論:成功制得5-FU直腸溫敏膠膜劑。
[Abstract]:Objective: to prepare 5-fluorouracil (5-Fu) rectal temperature sensitive adhesive. Methods: Poloxamer 407 (P407) and Poloxamer 188 (P188) were used as temperature-sensitive materials to prepare 5-FU rectal thermo-sensitive film. The mass concentration of 5-Fu and the kinds of biological adhesives were screened. Results: using P407-P188 (17: 2. 5) as temperature sensitive material, 2% carbomer 940 was used as biological adhesive and pH was 7.1. A temperature sensitive gel of 5 FU was prepared with a specification of 500 mg/30 ml. The gel was free flowing liquid at room temperature. The gel film was formed by phase transformation under physiological conditions. The phase transition temperature was 35 ~ 36.6 鈩,
