依達(dá)拉奉對大鼠慢性阻塞性肺病干預(yù)作用的研究

【摘要】:目的:研究自由基清除劑依達(dá)拉奉對慢性阻塞性肺。–OPD)大鼠肺損傷的作用,并探討其可能的作用機制,開拓依達(dá)拉奉對COPD的臨床應(yīng)用價值。 方法:健康雄性SD大鼠60只隨機分為6組,即空白對照組(空白組)、COPD模型組(COPD組)、N-乙酰半胱氨酸(NAC)陽性藥物對照組(NAC組)、依達(dá)拉奉低劑量組(Eda1組)、依達(dá)拉奉中劑量組(Eda2組)和依達(dá)拉奉高劑量組(Eda3組)。COPD模型組:參照宋一平脂多糖(LPS)氣管內(nèi)滴入加被動吸煙法(LPS+熏煙)復(fù)制COPD大鼠模型;NAC組:運用同樣的造模方法復(fù)制COPD大鼠模型,每次被動吸煙前予以NAC(20mg/kg)腹腔注射;依達(dá)拉奉低、中、高劑量組:造模方法同COPD模型組,每次熏煙前分別予以依達(dá)拉奉10mg/kg、20mg/kg、40mg/kg腹腔注射?瞻讓φ战M:不做熏煙處理,氣管滴注及腹腔注射均予以生理鹽水。造模4周后,測定大鼠肺功能,經(jīng)微機處理計算出呼吸頻率(f)、呼氣阻力(RI)、動態(tài)肺順應(yīng)性(Cdyn)及吸氣峰流速(PIF)。取血,檢測各組大鼠血漿丙二醛(MDA)水平和超氧化物歧化酶(SOD)活力。取大鼠支氣管肺泡灌洗液(BALF),進(jìn)行白細(xì)胞計總數(shù)、白細(xì)胞分類計數(shù),測定BALF上清液腫瘤壞死因子α(TNF-α)、白細(xì)胞介素8(IL-8)的水平。取右上肺做病理切片,HE染色,測量平均內(nèi)襯間隔(MLI)和平均肺泡數(shù)(MAN),免疫組化方法檢測各組大鼠肺組織中基質(zhì)金屬蛋白酶9(MMP-9)、基質(zhì)金屬蛋白酶12(MMP-12)、組織金屬蛋白酶抑制因子(TIMP-1)的表達(dá)。 結(jié)果:1. COPD組大鼠f、RI明顯高于空白組,差異有統(tǒng)計學(xué)意義(p0.05),而Cdyn、PIF顯著降低,與空白組有顯著性差異(p0.05),NAC與中、高劑量的依達(dá)拉奉可以顯著改善大鼠RI、Cdyn、PIF,差異有統(tǒng)計學(xué)意義(p0.05)。 2. COPD模型組大鼠血漿氧化產(chǎn)物MDA水平顯著升高,與空白組相比,差異有統(tǒng)計學(xué)意義(p0.05)。NAC與各劑量的的依達(dá)拉奉均能降低大鼠血漿MDA水平,差異有統(tǒng)計學(xué)意義(p0.05)。 3. COPD組大鼠白細(xì)胞總數(shù)與中性粒細(xì)胞百分率明顯增加,差異有統(tǒng)計學(xué)意義(p0.05)。NAC與中、高劑量的依達(dá)拉奉可以顯著降低白細(xì)胞總數(shù),差異有統(tǒng)計學(xué)意義(p0.05),中性粒細(xì)胞百分率可被NAC及高劑量的依達(dá)拉奉降低,差異有統(tǒng)計學(xué)意義(p0.05)。COPD組大鼠BALF中TNF-α水平顯著升高,差異有統(tǒng)計學(xué)意義(p0.05),NAC與各劑量的依達(dá)拉奉均可降低大鼠BALF中TNF-α水平,差異有統(tǒng)計學(xué)意義(p0.05)。 4.COPD組大鼠肺組織呈現(xiàn)肺泡融合、肺泡腔擴張等明顯肺氣腫樣病理改變,NAC組及Eda3組病理變化相對減輕。COPD組大鼠與空白組相比,平均內(nèi)襯間隔(MLI)明顯增加,平均肺泡數(shù)(MAN)顯著減少,差異均有統(tǒng)計學(xué)意義(p0.05)。NAC與中、高劑量的依達(dá)拉奉可以減小MLI,增加MAN,差異有統(tǒng)計學(xué)意義(p0.05)。 5. COPD組大鼠支氣管肺組織MMP-9、MMP-12及TIMP-1表達(dá)顯著增加,MMP-9/TIMP-1和MMP-12/TIMP-1比值增大,,與空白組相比差異均有統(tǒng)計學(xué)意義(p0.05)。NAC及中、高劑量的依達(dá)拉奉可以抑制MMP-9、MMP-12和TIMP-1的表達(dá),降低MMP-9/TIMP-1和MMP-12/TIMP-1比值,與COPD組相比,差異均具有統(tǒng)計學(xué)意義(p0.05)。 結(jié)論:大鼠造模過程中同時給予藥物干預(yù)可以延緩COPD的病理進(jìn)程,NAC和中、高劑量的依達(dá)拉奉可通過抗氧化,抑制炎癥細(xì)胞聚集,抑制炎癥因子釋放,降低蛋白酶的表達(dá),平衡蛋白酶/抗蛋白酶系統(tǒng)而發(fā)揮一定的抗COPD作用。
[Abstract]:Objective : To study the effect of free radical scavenging agent Edara on lung injury in rats with chronic obstructive pulmonary disease ( COPD ) , and to explore its possible mechanism of action and to explore the clinical application value of Edara in COPD .
Methods : 60 healthy male SD rats were randomly divided into six groups : blank control group ( blank group ) , COPD model group ( COPD group ) , N - acetyl cysteine ( NAC ) positive drug control group ( NAC group ) , Eda1 group ( Eda1 group ) , Eda2 group ( Eda2 group ) and Eda3 group ( Eda3 group ) .
NAC group : Using the same molding method to replicate the model of COPD rats , NAC ( 20 mg / kg ) was injected intraperitoneally before each passive smoking ;
The levels of plasma malondialdehyde ( MDA ) and superoxide dismutase ( SOD ) in lung tissues of rats were measured by computer . The expression of matrix metalloproteinase 9 ( MMP - 9 ) , matrix metalloproteinase 12 ( MMP - 12 ) and tissue inhibitor of metalloproteinase - 1 ( TIMP - 1 ) in lung tissues of rats were measured .
Results : 1 . In COPD group , f and RI were significantly higher than those in the blank group ( p < 0.05 ) , but there was a significant difference between the two groups ( p < 0.05 ) .
2 . Compared with blank group , the level of MDA in plasma oxidation products of COPD model group was significantly higher than that in blank group ( p < 0.05 ) .
3 . The total number of white blood cells and neutrophil percentage increased significantly in COPD group ( p < 0.05 ) . The difference was statistically significant ( p < 0.05 ) . The level of TNF - 偽 in BALF of COPD group was significantly increased ( p < 0.05 ) . The level of TNF - 偽 in BALF of COPD group was significantly decreased ( p < 0.05 ) .
4 . Compared with the blank group , the mean liner spacing ( MLI ) was significantly increased and the mean alveolar number ( MAN ) was significantly decreased compared with the blank group ( p 0.05 ) .
5 . The expression of MMP - 9 , MMP - 12 and TIMP - 1 in broncho - lung tissues of COPD group was significantly increased , and the ratio of MMP - 9 / TIMP - 1 and MMP - 12 / TIMP - 1 increased significantly ( p < 0.05 ) .
Conclusion : At the same time , drug intervention can delay the pathological process of COPD , NAC and middle and high doses of Edara can exert a certain anti - COPD effect by resisting oxidation , inhibiting inflammation cell aggregation , inhibiting the release of inflammatory factors , reducing the expression of protease , balancing the protease / protease system .
