甘草酸對(duì)高糖誘導(dǎo)的腎小球系膜細(xì)胞氧化應(yīng)激損傷的影響

【摘要】:目的探討甘草酸(GA)對(duì)高糖誘導(dǎo)的腎小球系膜細(xì)胞氧化應(yīng)激損傷的影響。方法將HBZY-1細(xì)胞分為:正常對(duì)照組(NG組)、高糖組(HG組)、高糖+GA組(HG+GA組)。采用四甲基偶氮唑藍(lán)(MTT)法檢測(cè)細(xì)胞增殖活性。用紫外分光光度法檢測(cè)超氧化物歧化酶(SOD)和丙二醛(MDA)水平,用激光共聚焦檢測(cè)活性氧(ROS)變化。采用免疫組織化學(xué)和Western blot檢測(cè)錳超氧化物歧化酶(Mn-SOD)蛋白的表達(dá),用Q-PCR檢測(cè)Mn-SOD mRNA。結(jié)果 (1)各組HBZY-1細(xì)胞形態(tài)變化:HBZY-1細(xì)胞為菱形,NG組細(xì)胞形態(tài)正常,結(jié)構(gòu)清晰可辨;HG+GA組細(xì)胞數(shù)量略增多,個(gè)別細(xì)胞胞體略肥大;HG組細(xì)胞結(jié)構(gòu)不甚清晰,細(xì)胞扁平,數(shù)量明顯增多,胞體略肥大。(2)各組HBZY-1細(xì)胞的增殖效應(yīng):與NG組相比,HG組OD值明顯升高(P0.05),HG+GA組與HG組相比OD值降低(P0.05)。(3)RDS含量:HG組RDS相對(duì)含量較NG組有所上升,HG+GA組ROS相對(duì)含量下降(P0.05)。(4)各組細(xì)胞中Mn-SOD的表達(dá):與NG組相比,HG組Mn-SOD相對(duì)表達(dá)減少(P0.05),HG+GA組與HG組相比Mn-SOD表達(dá)升高(P0.05)。結(jié)論 GA對(duì)高糖誘導(dǎo)的HBZY-1細(xì)胞異常增殖有一定的抑制作用,GA可通過(guò)氧化應(yīng)激保護(hù)高糖誘導(dǎo)腎小球系膜細(xì)胞所致的細(xì)胞肥大和細(xì)胞損傷,GA能調(diào)節(jié)高糖誘導(dǎo)下Mn-SOD的表達(dá)。
[Abstract]:Objective to investigate the effect of glycyrrhizic acid (GA) on oxidative stress injury induced by high glucose in glomerular Mesangial cells. Methods HBZY-1 cells were divided into three groups: normal control group (NG group), high glucose group (HG group) and high glucose group (GA group). Cell proliferation activity was detected by MTT assay. The levels of superoxide dismutase (SOD) and malondialdehyde (MDA) were detected by ultraviolet spectrophotometry, and the changes of reactive oxygen species (Ros) were detected by confocal laser. The expression of manganese superoxide dismutase (Mn-SOD) protein was detected by immunohistochemistry and Western blot, and Mn-SOD mRNA was detected by Q-PCR. Results 1) the morphological changes of HBZY-1 cells in each group were normal, the number of cells in the group of HGGA was slightly increased, the cell structure was not very clear, the number of cells was flat and the number of cells in HG group was obviously increased. Proliferation effect of HBZY-1 cells in each group: compared with NG group, the OD value of HG group was significantly higher than that of HG group. The OD value of HG group was significantly lower than that of HG group. The OD value of HG group was lower than that of HG group. The relative content of RDS in group 1: HG was higher than that in group NG. The expression of Mn-SOD: compared with NG group, the relative expression of Mn-SOD in HG group was lower than that in HG group. The expression of Mn-SOD in HG GA group was higher than that in HG group. Conclusion GA can inhibit the abnormal proliferation of HBZY-1 cells induced by high glucose. Ga can protect the cell hypertrophy and cell damage induced by high glucose in glomerular Mesangial cells by oxidative stress. The GA can regulate the expression of Mn-SOD induced by high glucose.
