二氫楊梅素對小鼠葡聚糖硫酸鈉結(jié)腸炎的干預(yù)作用

【摘要】:目的探討二氫楊梅素(DMY)對葡聚糖硫酸鈉(DSS)誘導(dǎo)的小鼠潰瘍性結(jié)腸炎(UC)的作用效果。方法將60只Balb/C小鼠隨機(jī)分為正常對照組、模型組、DMY低劑量組(50 mg·kg~(-1)·d~(-1))、DMY中劑量組(100 mg·kg~(-1)·d~(-1))、DMY高劑量組(200 mg·kg~(-1)·d~(-1))和5-氨基水楊酸組(50 mg·kg~(-1)·d~(-1)),各10只。正常對照組小鼠每日給予蒸餾水飲用,其余組小鼠給予4%DSS連續(xù)飲用7 d,誘導(dǎo)小鼠急性UC模型,與此同時(shí)給予對應(yīng)劑量的藥物灌胃治療。7 d后處死小鼠并計(jì)算小鼠疾病活動指數(shù)(DAI)、光鏡下結(jié)腸組織損傷評分(TDI),流式細(xì)胞法測定外周血中調(diào)節(jié)性T淋巴細(xì)胞(Treg)、輔助性T細(xì)胞17(Th17)占CD4+T細(xì)胞的百分比,Western blot及免疫組化法測定結(jié)腸組織中基質(zhì)金屬蛋白酶9(MMP 9)的表達(dá)水平。結(jié)果與正常對照組相比,模型組DAI、TDI明顯升高(P0.01),Treg/Th17比值明顯降低(P0.01),MMP 9表達(dá)升高(P0.05)。與模型組相比,DMY低、中、高劑量組小鼠Treg/Th17比值明顯升高(P0.01),MMP 9表達(dá)降低(P0.05),DMY中、高劑量組小鼠DAI、TDI明顯降低(P0.01),且隨著治療劑量增加,升高或降低幅度愈明顯。結(jié)論 DMY可以顯著緩解DSS誘導(dǎo)的小鼠UC炎癥,且與劑量呈正相關(guān)。
[Abstract]:Objective to investigate the effect of dihydromyricetin (DMY) on UC induced by dextran sodium sulfate (DSS) in mice with ulcerative colitis. Methods Sixty Balb/C mice were randomly divided into normal control group, model group with low dose of DMY (50 mg / kg / kg) / d ~ (-1), medium dose group (n = 10) and control group (n = 10), respectively, with a dose of 100 mg / d ~ (-1) / d ~ (-1) (n = 10) and a group of 5 ~ (-) aminosalicylic acid (n = 10), respectively, with a dose of 200 mg / kg ~ (-1) ~ (-1) ~ (-1) ~ (-1) d ~ (-1) ~ (-1) ~ (-1) (n = 10) and a group of 5 ~ (-) aminosalicylic acid group (n = 10). The mice in the control group were given distilled water daily and the other mice were given 4%DSS for 7 days to induce the acute UC model. At the same time, the mice were sacrificed after oral administration of the corresponding dose of drugs for 7 days, and the disease activity index (DAI) was calculated. The colonic tissue injury score (TDI) was evaluated under light microscope. The Tregn of regulatory T lymphocytes in peripheral blood was measured by flow cytometry. The expression of matrix metalloproteinase (9(MMP 9) in colon tissue was detected by Western blot and immunohistochemical method. Results compared with the normal control group, the TDI of the model group was significantly higher than that of the control group, and the ratio of Treg-Th17 to Treg-17 in the model group was significantly lower than that in the control group. The expression of MMP9 in the model group was significantly lower than that in the control group (P 0.05). Compared with the model group, the Treg/Th17 ratio in the high dose group was significantly higher than that in the model group, and the expression of MMP9 in the high dose group was significantly higher than that in the model group. In the high dose group, the expression of Treg/Th17 in the high dose group was significantly lower than that in the model group, and with the increase of the therapeutic dose, the increase or decrease was more obvious. Conclusion DMY can significantly relieve UC inflammation induced by DSS in mice and has a positive correlation with the dose.
