氯沙坦對(duì)大鼠廢用性骨骼肌不同肌球蛋白亞型基因表達(dá)的影響

【摘要】:目的探討氯沙坦對(duì)大鼠廢用性骨骼肌不同肌球蛋白亞型基因表達(dá)的影響。方法采用大鼠尾部懸吊2周建立廢用性肌肉萎縮模型,藥物干預(yù)組給予氯沙坦灌胃2周。采用實(shí)時(shí)熒光定量PCR測(cè)試比目魚肌中不同肌球蛋白亞型基因表達(dá)量,冰凍切片測(cè)試骨骼肌中毛細(xì)血管密度和毛細(xì)血管數(shù)/肌纖維數(shù)比值。結(jié)果 (1)與C組相比,TS組大鼠體重、比目魚肌重量和相對(duì)重量均降低(P0.05或P0.01),與TS組相比,TD組大鼠體重、比目魚肌重量和相對(duì)重量均升高(P0.05);(2)與C組相比,TS組、TD組大鼠比目魚肌毛細(xì)血管密度和毛細(xì)血管數(shù)/肌纖維數(shù)比值顯著下降(P0.05),TD組與TS組相比有顯著性增加(P0.05);(3)與C組相比,TS、TD組的MHC I mRNA、MHCⅡa mRNA、MHCⅡb mRNA表達(dá)量均降低(P0.01、P0.05、P0.05),TD組的MHC I mRNA表達(dá)量與TS組相比升高(P0.05)。結(jié)論大鼠尾部懸吊14天,比目魚肌出現(xiàn)明顯萎縮。服用氯沙坦可通過改善比目魚肌的血液循環(huán),提高M(jìn)HC I mRNA的表達(dá),具有一定的抑制骨骼肌萎縮作用。
[Abstract]:Objective to investigate the effect of losartan on gene expression of different myosin subtypes in rat skeletal muscle. Methods the rat model of disused muscular atrophy with tail suspension for 2 Zhou Jianli was used. Losartan was administered intragastrically for 2 weeks in the drug intervention group. The gene expression of different myosin subtypes in soleus muscle was measured by real-time fluorescence quantitative PCR, and the capillary density and the ratio of capillary number to muscle fiber number in skeletal muscle were measured by frozen sections. Results compared with group C, the body weight, soleus muscle weight and relative weight of TS group were decreased by P0.05 or P0.01, and those of TD group were significantly lower than those of TS group. Compared with group C, the density of capillaries and the ratio of capillaries to fiber number of soleus muscle in TD group decreased significantly. The ratio of capillary number to muscle fiber number in group TD was significantly higher than that in group T (P 0.05) and group C was significantly higher than that in group TS.
