星點設(shè)計-效應(yīng)面法優(yōu)化雙嘧達(dá)莫包合物制備工藝

【摘要】:目的:星點設(shè)計-效應(yīng)面法優(yōu)化雙嘧達(dá)莫(DIP)-β-環(huán)糊精(β-CD)包合物制備工藝。方法:以飽和水溶液法制備DIP-β-CD包合物,采用星點設(shè)計優(yōu)化制備工藝,以β-CD/DIP質(zhì)量比、包合時間和包合溫度為自變量,包合率為因變量,分別進行多元線性回歸和二項式方程擬合,優(yōu)選最佳處方工藝并進行驗證。結(jié)果:二項式方程擬合度較高(r2=0.911 3,P0.001),預(yù)測性好;效應(yīng)面法優(yōu)選最佳工藝條件為β-CD/DIP質(zhì)量比5.7,包合時間5.5 h,包合溫度50℃;最佳工藝驗證偏差均5%,所得包合物溶解度及釋藥速度均提高。結(jié)論:應(yīng)用星點設(shè)計-效應(yīng)面優(yōu)化法能夠精確有效地優(yōu)化DIP-β-CD包合物制備工藝,且最優(yōu)工藝穩(wěn)定可行,可用于工業(yè)生產(chǎn)。
[Abstract]:Objective: to optimize the preparation process of dipyridamole DIP- 尾-cyclodextrin (尾-CD) inclusion complex by star design-effect surface method. Methods: DIP- 尾 -CD inclusion complex was prepared by saturated aqueous solution method. The preparation process was optimized by star design. The 尾 -CD / dip mass ratio, inclusion time and inclusion temperature were taken as independent variables and the inclusion rate was used as dependent variable, and the multivariate linear regression and binomial equation were used to fit the DIP- 尾 -CD inclusion complex. Select the best prescription process and verify it. Results: the binomial equation had higher fitting degree and good predictability, and the optimum conditions were as follows: 尾 -CD- / dip ratio 5.7, inclusion time 5.5 h, inclusion temperature 50 鈩,
