環(huán)境中低濃度四溴雙酚A對顫蚓的毒性研究

【摘要】:本文以顫蚓為受試生物,研究四溴雙酚A(TBBPA)對顫蚓的毒性效應(yīng)以及顫蚓對TBBPA的富集效果。首先通過急性毒性實(shí)驗(yàn)測得TBBPA對顫蚓的96 h—LC50;其次根據(jù)急性毒性試驗(yàn)結(jié)果進(jìn)行半靜態(tài)試驗(yàn),選取0、0.025、0.05、0.25、0.50和1.0 mg·L-1六組濃度進(jìn)行染毒實(shí)驗(yàn),再選取1 mg·L-1濃度培養(yǎng)1、3、5、7和10 d后,測定各組顫蚓體內(nèi)酶活性;最后設(shè)置TBBPA高濃度(1 mg·L-1)和低濃度(0.2 mg·L-1)兩組染毒7 d后,測定顫蚓對TBBPA的富集系數(shù)(BCF)。主要結(jié)果如下:1.隨著暴露濃度和暴露時(shí)間的增加,各處理組顫蚓的死亡率上升,TBBPA毒性明顯增強(qiáng),存在明顯劑量—效應(yīng)關(guān)系。TBBPA對顫蚓48 h、72 h、96 h的LC50分別為11.295 mg·L-1、3.864 mg·L-1、2.204 mg·L-1,安全濃度為0.22 mg·L-1。2.暴露1 d和7 d后,顫蚓體內(nèi)的超氧化物岐化酶(SOD)表現(xiàn)為誘導(dǎo)效應(yīng),其中0.05 mg·L-1濃度組誘導(dǎo)最大(P0.01),分別為對照組的1.36和1.61倍;過氧化氫酶(CAT)活性以誘導(dǎo)效應(yīng)為主,同樣在0.05 mg·L-1濃度組達(dá)到峰值(P0.01),但暴露7 d的CAT活性總體低于暴露1 d的CAT活性;過氧化物酶(POD)活性呈現(xiàn)先誘導(dǎo)后抑制的變化趨勢,在0.025 mg·L-1濃度組即達(dá)到最大誘導(dǎo)(P0.01),分別為對照組的1.28和1.38倍,在1 mg·L-1濃度組時(shí)活性降至最低,抑制率分別為8%和20%;乙酰膽堿酯酶(AChE)活性先增加后降低,各處理組AChE活性均顯著高于對照組(P0.01),當(dāng)TBBPA濃度為0.25 mg·L-1時(shí)達(dá)到峰值,分別為對照組的1.25和1.20倍。3.在1 mg·L-1TBBPA暴露的10 d內(nèi),SOD、CAT和AChE活性均出現(xiàn)先升高后下降的變化趨勢,其活性分別在第3 d和第5 d達(dá)到最大值(P0.01)。POD的變化則分為降低、升高、再降低3個(gè)階段,其活性在第3 d到達(dá)峰值(P0.01)。4.隨著染毒液濃度的升高,水體中殘留和顫蚓體內(nèi)累積的TBBPA含量均逐漸增加,其中高濃度組(1 mg·L-1)BCF值高于低濃度組(0.2 mg·L-1),兩組平均BCF值為31.55,表明顫蚓對TBBPA的富集并不顯著。
[Abstract]:In this paper, the toxic effects of tetrabromobisphenol A (TBBPA) on vermis and the enrichment effect of vermis on TBBPA were studied. First, 96 h-LC50 of TBBPA was measured by acute toxicity test, secondly, semi-static test was carried out according to the results of acute toxicity test. Six groups of 0 0. 025, 0. 25, 0. 25 and 1. 0 mg L ~ (-1) were selected for exposure test, and then 1 mg L ~ (-1) was cultured for 1 ~ (3) C ~ (5) for 7 and 10 days, and then 1 mg 路L ~ (-1) was cultured for 7 and 10 days. The enzyme activity in each group was determined, and the enrichment factor of TBBPA was determined after 7 days of exposure to TBBPA at high concentration (1 mg / L) and low concentration (0. 2 mg / L). The main results are as follows: 1. With the increase of exposure concentration and exposure time, the death rate of vermis increased and the toxicity of TBBPA was significantly increased. The LC50 of 48 h / 72 h / 96 h was 11.295 mg / L ~ (3.864) mg / L ~ (-1) and 0.22 mg / L ~ (-1), respectively, and the safe concentration was 0.22 mg / L ~ (-1). After exposure for 1 d and 7 d, the superoxide dismutase (SOD) in the vermis was induced, and the maximal induction was found in the 0.05 mg L-1 group, which was 1.36 and 1.61 times higher than that in the control group, and the activity of catalase CAT was mainly induced. The activity of CAT was lower than that of CAT at 7 days of exposure, and the activity of peroxidase (POD) showed the tendency of first induction and then inhibition, and the activity of P0. 01 and P0. 01% reached the peak value in the group of 0. 05 mg L-1, but the activity of CAT was lower than that of 1 day of exposure. In the 0.025 mg L-1 group, the maximum induction P0.01D was 1.28 and 1.38 times of that in the control group, and the activity decreased to the lowest in the 1 mg L-1 group, and the inhibition rates were 8% and 20%, respectively, and the acetylcholinesterase (ache) activity increased first and then decreased. The activity of AChE in each treatment group was significantly higher than that in the control group (P 0.01), and reached the peak value when the concentration of TBBPA was 0.25 mg L-1, which was 1.25 times and 1.20 times as much as that of the control group, respectively. The activities of cat and AChE increased first and then decreased within 10 days after exposure to 1 mg L-1TBBPA. The changes of cat and AChE activities reached the maximum on the 3rd and 5th day, respectively, and were decreased, increased, and then decreased. Its activity reached its peak value on the 3rd day (P0.01U. 4). With the increase of the concentration of venom, the TBBPA content of residual and vermis accumulated in water increased gradually, and the value of 1 mg L-1)BCF in the high concentration group was higher than that in the low concentration group, and the average BCF value of the two groups was 31.55, which indicated that the accumulation of TBBPA by the vermis was not significant.
