近紅外光介導(dǎo)的尺寸可逆轉(zhuǎn)變超聲造影劑的制備及其性質(zhì)

【摘要】:制備一種納米級(jí)的載藥超聲造影劑,該納米粒可有效累積分布至腫瘤深層部位,且體外實(shí)驗(yàn)證明,在升高溫度或近紅外光輻照下可實(shí)現(xiàn)納米粒尺寸的納米-微米轉(zhuǎn)變:評(píng)價(jià)其體外粒徑變化規(guī)律、體外超聲顯像效果及體內(nèi)分布情況。所制備的納米粒呈球形,脂質(zhì)膜中可見中空金納米球(hollow gold nanoparticle,HAu NS)的負(fù)載,粒徑為302±5 nm,分散性指數(shù)為0.195±0.018,分布較均勻。在近紅外光輻照(1或2 W·cm~(-2))下導(dǎo)致含有該納米粒的溶液(以HAu NS計(jì),0.2、0.04和0.02 g·L~(-1))升溫迅速,利用激光粒度儀的控溫功能,發(fā)現(xiàn)升溫至52℃左右時(shí)即可檢測(cè)到一定量的微米級(jí)粒子產(chǎn)生;在體外超聲顯像實(shí)驗(yàn)中,納米粒經(jīng)近紅外光輻照后超聲診斷儀檢測(cè)到大量上揚(yáng)的微泡,可增強(qiáng)體外超聲顯像的效果;體內(nèi)分布實(shí)驗(yàn)表明,該納米?梢蚰[瘤的高滲透及滯留效應(yīng)(EPR效應(yīng))在腫瘤中大量累積。本研究制備了可在腫瘤內(nèi)部大量累積的納米-微米可逆轉(zhuǎn)變的超聲顯像納米粒,為實(shí)現(xiàn)腫瘤病灶部位的超聲顯像同時(shí)結(jié)合光熱-化學(xué)治療提供了一種可行的方案。
[Abstract]:A kind of nano-scale drug-loaded ultrasound contrast agent was prepared, which can effectively accumulate and distribute to the deep part of the tumor, and has been proved by experiments in vitro. The nanometer-micron transition of nanoparticles can be realized at elevated temperature or near infrared irradiation. The change of particle size in vitro, the effect of in vitro ultrasound imaging and the distribution in vivo are evaluated. The prepared nanoparticles were spherical, and the hollow gold nanospheres hollow gold nanoparticlehau NSN could be seen in the lipid membrane. The particle size was 302 鹵5 nm, the dispersion index was 0.195 鹵0.018, and the distribution was uniform. The solution containing the nanoparticles (0.2n 0.04 and 0.02g / L ~ (-1) ~ (-1) was induced by near-infrared irradiation (1 or 2 W / cm ~ (-1).
