透明質酸修飾的尿酸酶多囊脂質體體外穩(wěn)定性初步研究

【摘要】:目的:制備了透明質酸修飾的尿酸酶多囊脂質體(hyaluronic acid-uricase liposomes,UHMVL),并且對尿酸酶(uricase,UC)以及UHMVL在體外的活性和穩(wěn)定性進行了初步的研究。方法:采用復乳法制備了UHMVL,并且測定了其包封率。然后從最適溫度(n=3)、最適p H(n=3)、體外穩(wěn)定性(n=3)初步考察了UHMVL和UC的差異。結果:UHMVL的包封率為(62.48±3.87)%。UHMVL及UC最適溫度均為40℃;UHMVL的最適p H值為8.0,UC的最適p H值為8.5。穩(wěn)定性測定結果表明,UHMVL體外穩(wěn)定性均比UC好。UHMVL在提高了UC在體外活性的同時,也明顯地增強了UC在體外的穩(wěn)定性。結論:UHMVL在提高了UC體外活性的同時,也明顯地增強了游離酶UC在體外的穩(wěn)定性。
[Abstract]:Aim: to prepare hyaluronic acid-uricase liposome UHMVLA modified by hyaluronidase, and to study the activity and stability of uricase (UC) and UHMVL in vitro. Methods: UHMVLs were prepared by double emulsion method and their encapsulation efficiency was determined. Then, the difference between UHMVL and UC was preliminarily investigated from the optimum temperature, the optimum temperature, the optimum pH, the in vitro stability and the in vitro stability.
