【摘要】：布格呋喃(AF-5)是已報道的一類具有抗焦慮活性的精神治療藥物之一,其結(jié)構(gòu)由一個沉香呋喃環(huán)骨架和一個正丁基側(cè)鏈組成。臨床藥代研究發(fā)現(xiàn)其新的代謝產(chǎn)物,推測其結(jié)構(gòu)可能為烷烴鏈端基的羰酸化產(chǎn)物。為了確證其結(jié)構(gòu),以中間體(4a R,7R,8a R)-8a-羥基-7-(2-羥丙基-2-基)-4a-甲基八氫萘-2(1氫)-酮為原料,經(jīng)6步反應,成功合成了目標化合物,總收率16.7%�；衔锏慕Y(jié)構(gòu)均經(jīng)NMR和MS確證。
[Abstract]:Bugfuran (AF-5) is one of the reported psychotherapeutic drugs with antianxiety activity. Its structure consists of a cytoskeleton and a positive side chain of Ding Ji. New metabolites were found in clinical pharmacokinetic studies, which suggested that their structures might be carbonylation products of alkane chain terminal groups. In order to confirm its structure, the target compound was successfully synthesized from the intermediate 4a Rn 7RN 8a RZ-8a- hydroxypropyl -2-hydroxypropyl -4a- methyloctachloronaphthalene -2H _ 2H _ 2N _ 1 through six steps reaction, with the overall yield of 16.7b. The structures of the compounds were confirmed by NMR and MS.
