載藥熒光納米粒子的制備及在乳腺癌MCF-7細胞系的應(yīng)用及效果評價

【摘要】:利用兩親性聚乙二醇-聚乳酸共聚物(PEG-PDLLA)包覆熒光染料(DPBA)和紫杉醇(PTX),通過自組裝方法制得載藥熒光納米粒子DPBA/PTX@PEG-PDLLA.納米粒子尺寸均一,具有良好的生物相容性.對納米粒子的發(fā)光性質(zhì)、載藥量和體外藥物釋放等進行了表征,并考察了納米粒子對乳腺癌細胞MCF-7的抑制效果,觀察了MCF-7細胞對納米粒子的攝取情況.結(jié)果表明,DPBA/PTX@PEG-PDLLA納米粒子具有較強的紅光發(fā)射,不僅可以用于MCF-7腫瘤細胞質(zhì)熒光成像,而且對腫瘤細胞的增殖具有一定的抑制能力.
[Abstract]:The fluorescent dye DPBA and paclitaxel were coated with amphiphilic poly (ethylene glycol) polylactic acid copolymer (PEG-PDLLA). The fluorescent nanoparticles DPBA / PTXPEG-PDLLAA were prepared by self-assembly method. The nanoparticles are homogeneous in size and have good biocompatibility. The luminescence properties, drug loading and drug release in vitro of nanoparticles were characterized. The inhibitory effect of nanoparticles on MCF-7 of breast cancer cells was investigated, and the uptake of nanoparticles in MCF-7 cells was observed. The results showed that DPBA / PTX @ PEG-PDLLA nanoparticles had strong red light emission, which could not only be used for fluorescence imaging of MCF-7 tumor cytoplasm, but also could inhibit the proliferation of tumor cells.
