重組人卵泡刺激素類似物對幼齡小鼠卵巢發(fā)育的影響

【摘要】:目的:探討重組人卵泡刺激素(recombinant follicle stimulating hormone,r FSH)類似物對幼齡小鼠卵巢發(fā)育的影響。方法:3周齡小鼠隨機分為6組:溶媒空白對照組、果納芬單次注射組(0.5 IU)和果納芬3次注射組(0.5 IU·d-1,連續(xù)給藥3 d);r FSH類似物低劑量組(0.375 IU)、中劑量組(0.75 IU)和高劑量組(1.5 IU)均在停藥24 h后摘取卵巢稱重并計算卵巢指數(shù),卵巢組織經(jīng)HE染色,觀察各級卵泡所占比例。結(jié)果:與空白對照組相比,r FSH類似物能劑量依賴性地促進卵巢增重,顯著增加卵巢指數(shù)(P0.05)。0.375 IU劑量即可顯著促進幼齡小鼠卵巢指數(shù)的增加(P0.05),1.5 IU劑量組較陽性藥物果納芬3次注射組無顯著性差異;0.75 IU r FSH類似物較空白對照組可顯著降低原始卵泡所占比例,提高成熟卵泡和黃體所占比例(P0.01),且1.5 IU劑量組較陽性藥物果納芬3次注射組能更有效地降低原始卵泡的比例,提高成熟卵泡和黃體所占比例(P0.01)。結(jié)論:r FSH類似物不僅能夠促進幼齡小鼠卵巢增重以及促進原始卵泡、初級卵泡和次級卵泡的發(fā)育,而且具長效性,即單次注射即可使血藥濃度維持3 d。
[Abstract]:Aim: to investigate the effects of recombinant follicle stimulating hormoney analogue on ovarian development in young mice. Methods Twenty three week-old mice were randomly divided into 6 groups: solvent blank control group. In the single injection group (0.5 IUU) and the 3 times injection group (0.5 IU d-1), the low dose group (0.375 IUU) and the middle dose group (0.75 IUU) and the high-dose group (1.5-IUU) were taken out 24 hours after the withdrawal of the drug and the ovarian index was calculated. The proportion of follicles was observed by HE staining in ovarian tissues. Results: compared with the control group, the analogue of r FSH could promote ovarian weight gain in a dose-dependent manner. The ovarian index of young mice was significantly increased by increasing the dosage of P0.05U. 0.375IU. The dose of P0.05U 1.5IU was significantly lower than that of the three injection group of positive drug, the analogue of 0.75IU r FSH was significantly lower than that of the blank control group. Proportion of low primordial follicles, The proportion of mature follicles and corpus luteum was increased, and the proportion of mature follicles and corpus luteum was increased more effectively in 1.5 IU group than that in the three injection groups of positive drug Funafen, and the proportion of mature follicles and corpus luteum was increased. Conclusion the fraction r FSH analogue can not only promote ovarian weight gain and the development of primordial follicles, primary follicles and secondary follicles in young mice, but also have a long-term effect, that is, a single injection can maintain the blood concentration for 3 days.
