寧夏枸杞內(nèi)生菌的抗菌和抗腫瘤活性研究

【摘要】:為篩選具有抗菌抗腫瘤活性的枸杞內(nèi)生菌資源,對分離自寧夏枸杞的10株內(nèi)生菌發(fā)酵液的抑菌活性和體外細(xì)胞毒活性進(jìn)行檢測,通過ITS和16S rDNA序列分析對菌株進(jìn)行了分子鑒定。結(jié)果表明,5株內(nèi)生真菌對供試病原菌都未表現(xiàn)出抑制活性,放線菌AL6對3種病原真菌都有一定的抑制作用,而AL5僅對金黃色葡萄球菌有較強抑制活性。但內(nèi)生真菌的抗腫瘤活性明顯強于放線菌。當(dāng)發(fā)酵液濃度為0.2 g·L~(-1)時,有4株內(nèi)生真菌至少對1種腫瘤細(xì)胞的抑制率高于50%。序列分析表明,5株內(nèi)生真菌隸屬于曲霉屬、青霉屬和翹孢霉屬。5株內(nèi)生放線菌隸屬于鏈霉菌屬、微球菌屬和擬諾卡氏菌屬。曲霉屬菌株FL1對A549和HeLa細(xì)胞的抑制活性都較強,其IC_(50)分別為0.022,0.028 g·L~(-1),值得進(jìn)一步研究。
[Abstract]:In order to screen endophytic resources of Lycium barbarum with antibacterial and antitumor activities, the antibacterial activity and cytotoxic activity of 10 strains of endophytic bacteria from Ningxia Lycium barbarum were detected. The strains were identified by ITS and 16s rDNA sequence analysis.The results showed that 5 strains of endophytic fungi had no inhibitory activity against the pathogens tested, while actinomycetes AL6 had a certain inhibitory effect on the three pathogenic fungi, while AL5 only had strong inhibitory activity on Staphylococcus aureus.But the antitumor activity of endophytic fungi was obviously stronger than that of actinomycetes.When the fermentation broth concentration was 0.2 g / L ~ (-1), the inhibition rate of 4 strains of endophytic fungi on at least one tumor cell was higher than 50%.Sequence analysis showed that 5 endophytic fungi belong to Aspergillus, and 5 endophytic actinomycetes belong to Streptomyces, Micrococcus and Nocardia.The inhibitory activity of Aspergillus strain FL1 on A549 and HeLa cells was strong, and its IC _ (50) was 0.022 ~ 0.028 g / L ~ (-1), respectively, which was worthy of further study.
