他克莫司通過下調(diào)NFATc2抑制大鼠球囊損傷后頸總動脈內(nèi)膜增生

【摘要】:目的觀察他克莫司對大鼠球囊損傷后血管壁內(nèi)膜增生的干預(yù)作用并探討其機(jī)制。方法將27只健康雄性SD大鼠隨機(jī)分成假手術(shù)組、球囊損傷模型組及他克莫司組(n=9),除假手術(shù)組外,其余大鼠建立頸總動脈球囊損傷模型。他克莫司組大鼠灌胃給予他克莫司(1 mg·kg~(-1)),假手術(shù)組和球囊損傷模型組大鼠灌胃給予相同體積的雙蒸水,每日1次,連續(xù)給藥14 d。最后一次給藥后24 h,取損傷側(cè)血管行HE染色,計算管腔面積、血管內(nèi)膜面積(NIA)及內(nèi)膜面積/中膜面積(NIA/MA);real-time PCR法檢測腫瘤壞死因子(TNF)-α和白細(xì)胞介素(IL)~(-1)βm RNA表達(dá),免疫組化法檢測增殖細(xì)胞核抗原(PCNA)、活化T細(xì)胞核因子(NFAT)c2蛋白在血管壁中的表達(dá)。結(jié)果與假手術(shù)組相比,球囊損傷模型組管腔面積明顯減小,NIA、NIA/MA增加,TNF-α和IL~(-1)βm RNA表達(dá)增高,PCNA、NFATc2蛋白表達(dá)增高(P0.01)。與球囊損傷模型組相比,他克莫司組管腔面積增加,NIA增厚程度減輕,NIA/MA減小,TNF-α和IL~(-1)βm RNA的表達(dá)降低,PCNA、NFATc2蛋白的表達(dá)降低(P0.05或P0.01)。結(jié)論他克莫司可明顯抑制血管內(nèi)膜過度增生,其機(jī)制可能與其下調(diào)血管壁NFATc2蛋白和TNF-α、IL~(-1)βm RNA表達(dá)有關(guān)。
[Abstract]:Objective to investigate the effect of tacrolimus on intimal hyperplasia after balloon injury in rats.Methods 27 healthy male Sprague-Dawley rats were randomly divided into sham operation group, balloon injury model group and tacrolimus group.The rats in tacrolimus group were given tacrolimus 1 mg / kg, and rats in sham operation group and balloon injury model group were given the same volume of double distilled water once a day for 14 days.Immunohistochemical method was used to detect the expression of proliferating cell nuclear antigen (PCNA) and activated T cell nuclear factor (T) -NFATC _ 2 protein in vascular wall.Compared with the balloon injury model group, the lumen area of tacrolimus group increased and the thickness of NIA increased. NIA / MA decreased the expression of TNF- 偽 and ILF-1) 尾 m RNA. The expression of PCNANFATc2 protein decreased (P0.05) or P0.01 (P < 0.05).Conclusion tacrolimus can significantly inhibit intimal hypertrophy, which may be related to its down-regulation of the expression of NFATc2 protein and TNF- 偽 -l) 尾 m RNA.
