葉黃素的生物學作用及制劑研究進展

【摘要】:目的:為葉黃素的制劑研究提供參考。方法:以"葉黃素""藥理作用""劑型""應用""Lutein""Antioxidant"等為關(guān)鍵詞,組合查詢2006年1月-2016年7月在Pub Med、Elsevier、中國知網(wǎng)、萬方、維普等數(shù)據(jù)庫中的相關(guān)文獻,對葉黃素的理化性質(zhì)、體內(nèi)過程、抗氧化活性、生物學作用、劑型研究及應用現(xiàn)狀等方面進行綜述。結(jié)果與結(jié)論:共檢索到相關(guān)文獻696篇,其中有效文獻47篇。葉黃素含有獨特的紫羅酮環(huán)二羥基結(jié)構(gòu),可作為強抗氧化劑淬滅單線態(tài)氧和藍光過濾器,具有抗氧化、抗癌、保護視網(wǎng)膜、預防心血管疾病等生物學作用。但葉黃素的理化性質(zhì)不穩(wěn)定,在制劑過程中存在水溶性差、性質(zhì)不穩(wěn)定和生物利用度低等問題;葉黃素在體內(nèi)只能與脂類結(jié)合吸收,這一特點大大制約了其藥效的發(fā)揮。通過物理包埋和化學改性(如酯化反應)等方法改變劑型,并利用超微粉碎和微囊化等技術(shù)將葉黃素制備成油懸浮液、水分散性干粉、微膠囊和脂質(zhì)體,能較好地解決這些問題。但如何進一步提高葉黃素的生物利用度、減小對胃腸道作用的影響、改善傳統(tǒng)制劑粒徑過大等問題還有待進一步研究。
[Abstract]:Objective: to provide reference for the study of lutein preparation.Methods: using "lutein" "pharmacological action" "dosage form" "Lutein" and "Antioxidant" as the key words, the authors combined and inquired about the physical and chemical properties and in vivo process of lutein in Pub Medsite Elsevier, China Zhiwang, Wanfang, Weip and other databases from January 2006 to July 2016.The antioxidant activity, biological action, dosage form research and application status are reviewed.Results & conclusion: a total of 696 related literatures were retrieved, of which 47 were valid.Lutein has a unique structure of azolone ring dihydroxyl group and can be used as a powerful antioxidant to quench singlet oxygen and blue light filters. Lutein has many biological functions such as antioxidation anticancer protection of retina and prevention of cardiovascular diseases.However, the physicochemical properties of lutein are unstable, the water solubility is poor, the property is unstable and the bioavailability is low. Lutein can only be absorbed by lipids in vivo, which greatly restricts its pharmacodynamics.The preparation of lutein into oil suspension, water dispersible dry powder, microcapsule and liposome was prepared by physical entrapment and chemical modification (such as esterification reaction), and the preparation of lutein into oil suspensions, water dispersible dry powders, microcapsules and liposomes by ultramicro pulverization and microencapsulation.Can solve these problems well.However, how to further improve the bioavailability of lutein, reduce the effect on gastrointestinal tract, and improve the particle size of traditional preparations need to be further studied.
