活性維生素D3對(duì)進(jìn)展性腎炎大鼠腎小管間質(zhì)損傷的干預(yù)作用

【摘要】:目的探討活性維生素D3對(duì)進(jìn)展性腎炎大鼠腎小管間質(zhì)損傷的干預(yù)作用及對(duì)腎間質(zhì)纖維化的影響。方法實(shí)驗(yàn)動(dòng)物按數(shù)字隨機(jī)表法分為假手術(shù)組、進(jìn)展性腎炎組(模型組)、活性維生素D3干預(yù)組(治療組),每組7只。除對(duì)照組外,均采用右腎切除并兩次注射Ig G(OX-7)鼠抗人Thy1.1單克隆抗體制備進(jìn)展性腎炎大鼠模型;钚跃S生素D3干預(yù)8 w后,檢測(cè)各組大鼠24 h尿蛋白、血肌酐(Scr)、尿素氮(BUN)及電解質(zhì)的變化,并觀察腎組織病理改變。免疫熒光檢測(cè)腎小管間質(zhì)低氧誘導(dǎo)因子(HIF)-1α的表達(dá),原位雜交檢測(cè)腎小管間質(zhì)結(jié)締組織生長(zhǎng)因子(CTGF)mRNA的表達(dá)。結(jié)果與模型組比較,活性維生素D3治療不僅使HIF-1α、CTGF表達(dá)顯著下調(diào)(P0.05),還能使大鼠24 h尿蛋白、BUN明顯降低(P0.05),腎組織病理?yè)p害明顯減輕。結(jié)論慢性低氧上調(diào)了HIF-1α及前纖維因子的表達(dá),活性維生素D3可能通過(guò)明顯降低進(jìn)展性腎炎大鼠腎小管間質(zhì)HIF-1α、CTGF的表達(dá),減輕腎小管間質(zhì)纖維化而發(fā)揮腎臟保護(hù)作用。
[Abstract]:Objective to investigate the effect of active vitamin D 3 on renal tubulointerstitial injury and renal interstitial fibrosis in rats with progressive nephritis.Methods Experimental animals were randomly divided into sham-operated group, progressive nephritis group (model group) and active vitamin D3 intervention group (7 rats in each group).Except the control group, the rat model of progressive nephritis was established by right nephrectomy and twice injection of Ig GnOX-7) mouse monoclonal antibody against human Thy1.1.After intervention with active vitamin D _ 3 for 8 weeks, the changes of urine protein, creatinine creatinine, bun (bun) and electrolytes in 24 h were detected, and the pathological changes of renal tissue were observed.The expression of tubulointerstitial hypoxia inducible factor (HIF-1 偽) and tubulointerstitial connective tissue growth factor (CTGFN) mRNA were detected by immunofluorescence and in situ hybridization respectively.Results compared with the model group, the active vitamin D _ 3 treatment not only significantly decreased the expression of HIF-1 偽 -CTGF, but also significantly decreased the urine protein bun at 24 h and the pathological damage of renal tissue.Conclusion chronic hypoxia up-regulates the expression of HIF-1 偽 and prefibroin, and active vitamin D 3 may play a role in renal protection by significantly reducing the expression of HIF-1 偽 and CTGF in renal tubulointerstitial of rats with progressive glomerulonephritis.
