樺木酸納米粒的制備與表征

【摘要】:目的:制備樺木酸納米粒,并對(duì)其進(jìn)行表征。方法:以乙醇為溶劑、水為反溶劑,采用反溶劑重結(jié)晶法制備樺木酸納米粒。以粒徑為指標(biāo),采用單因素試驗(yàn)與正交試驗(yàn)優(yōu)化樺木酸納米粒處方工藝中樺木酸溶液質(zhì)量濃度、反溶劑-溶劑體積比、反溶劑滴加速度、反應(yīng)溫度和攪拌速度,并進(jìn)行驗(yàn)證試驗(yàn)。采用掃描電子顯微鏡、激光粒度儀、傅里葉紅外光譜儀和質(zhì)譜分析儀對(duì)所制得的樺木酸納米粒進(jìn)行表征。結(jié)果:最優(yōu)處方工藝為樺木酸溶液質(zhì)量濃度為3 mg/mL、反溶劑-溶劑體積比為1:1、反溶劑滴加速度為8 mL/min、反應(yīng)溫度為20℃、攪拌速度為900 r/min;所制樺木酸納米混懸液粒徑為(156.0±8.6)nm(n=3),凍干后粒徑為(235.0±12.2)nm(n=3),外觀近球形、大小均勻、形態(tài)較規(guī)整;與樺木酸原料藥比較,所制樺木酸納米粒的化學(xué)結(jié)構(gòu)沒(méi)有發(fā)生改變,分子量和質(zhì)荷比無(wú)明顯變化。結(jié)論:成功制得樺木酸納米粒。
[Abstract]:Objective: to prepare and characterize betulinic acid nanoparticles. Methods: birch acid nanoparticles were prepared by anti-solvent recrystallization with ethanol as solvent and water as antisolvent. Single factor test and orthogonal test were used to optimize the concentration of betulinic acid solution, the volume ratio of antisolvent to solvent, the acceleration of antisolvent drop, the reaction temperature and the stirring speed in the formulation process of birch acid nanoparticles. Using scanning electron microscope, laser particle size meter, Fourier transform infrared spectroscopy (FTIR) and mass spectrometer (MS) were used to characterize the prepared birch acid nanoparticles. Results: the optimum formulation conditions were as follows: the concentration of betulinic acid solution was 3 mg / mL, the volume ratio of antisolvent to solvent was 1: 1, and the acceleration of antisolvent drop was 1: 1.
