吉非替尼對斑馬魚胚胎發(fā)育及腫瘤耐藥基因表達的影響

【摘要】:目的研究吉非替尼對斑馬魚早期胚胎發(fā)育及abcb4腫瘤耐藥基因表達的影響。方法實驗動物采用斑馬魚,實驗分為吉非替尼組、阿霉素和吉非替尼的混合液組、空白對照組。將吉非替尼稀釋成不同濃度梯度的藥液,分別處理受精后0.5~1.5h(0.5~1.5hpf)的斑馬魚胚胎,觀察胚胎和幼魚的死亡率、孵化率及畸形率變化。統(tǒng)計各濃度不同時間點的胚胎死亡率,計算出IC_(50)的數(shù)值;統(tǒng)計48hpf和72hpf的胚胎孵化率及120hpf的畸形率;收集不同組別的斑馬魚胚胎,通過實時熒光定量PCR,檢測斑馬魚胚胎中abcb4基因的表達量。結(jié)果計算出吉非替尼在斑馬魚胚胎中的IC_(50)為16.18μmol/L。與對照組比較,高劑量吉非替尼(≥20μmol/L)顯著抑制胚胎的孵化,并有明顯的胚胎致死效應(yīng)和致畸效應(yīng);吉非替尼組的斑馬魚胚胎abcb4基因的mRNA水平變化不顯著,而阿霉素和吉非替尼的混合液組的abcb4基因的mRNA水平有明顯變化(P0.05)。結(jié)論吉非替尼對斑馬魚早期胚胎中abcb4腫瘤耐藥基因的表達水平無明顯影響(P0.05),但是吉非替尼能逆轉(zhuǎn)易耐藥藥物阿霉素的耐藥作用,提示使用斑馬魚可以構(gòu)建腫瘤耐藥模型。
[Abstract]:Objective to study the effects of gefitinib on the development of early embryos and the expression of drug-resistant genes in abcb4 tumor of zebrafish. Methods zebrafish were divided into three groups: gifitinib group, doxorubicin and gefitinib mixture group.
