【摘要】：目的建立鑒定頭孢特侖新戊酯中有關(guān)物質(zhì)的液相色譜-質(zhì)譜聯(lián)用分析方法,測(cè)定頭孢特侖新戊酯原料藥和制劑產(chǎn)品中的有關(guān)物質(zhì)。方法采用Syncronis C18(250 mm×4.6mm,5μm)色譜柱,10mmol·L-1乙酸銨-乙腈(體積比50∶50)作為流動(dòng)相等度洗脫分離;采集各有關(guān)物質(zhì)的UV譜、一級(jí)和二級(jí)質(zhì)譜圖,并根據(jù)所得信息對(duì)有關(guān)物質(zhì)進(jìn)行結(jié)構(gòu)分析。結(jié)果頭孢特侖新戊酯原料藥中檢測(cè)到8種有關(guān)物質(zhì),分別為6種合成副產(chǎn)物、1種水解產(chǎn)物和1種降解產(chǎn)物;制劑產(chǎn)品中除原料藥中的8種有關(guān)物質(zhì)外,還檢測(cè)到4種新的有關(guān)物質(zhì),分別為2種氧化產(chǎn)物、1種水解產(chǎn)物和1種降解產(chǎn)物。結(jié)論該方法靈敏度高,全面系統(tǒng)的闡述了頭孢特侖新戊酯中的有關(guān)物質(zhì),為其質(zhì)量控制和工藝優(yōu)化提供了參考依據(jù)。
[Abstract]:Objective to establish a liquid chromatography-mass spectrometry (LC-MS) method for the determination of related substances in ceftrientyl ester. Determination of related substances in ceftrientyl ester raw material and preparation products. Methods Syncronis C18250 mm 脳 4.6 mm ~ 5 渭 m column was used to elucidate 10 mmol 路L ~ (-1) ammonium acetate and acetonitrile (volume ratio 50: 50), and UV spectra of each related substance were collected. Results eight related substances were detected in ceftrientyl ester raw material, including 6 synthetic by-products, 1 hydrolytic product and 1 degradation product, respectively. In addition to 8 kinds of related substances in raw materials, four new related substances were detected in the preparation products, which were two oxidation products, one hydrolytic product and one degradation product, respectively. Conclusion the method has high sensitivity. The related substances in ceftrientyl ester are described systematically, which provides a reference for quality control and process optimization.
