【摘要】：目的:探討重組人Ⅱ型腫瘤壞死因子受體-抗體融合蛋白(rhTNFR∶Fc)對膠原誘導(dǎo)性關(guān)節(jié)炎(CIA)模型大鼠關(guān)節(jié)滑膜細(xì)胞NF-κB信號表達(dá)的影響。方法:將40只6周齡的雌性Wistar大鼠按照隨機(jī)數(shù)表法隨機(jī)分為空白組、模型組、醋酸潑尼松對照組和rhTNFR∶Fc治療組,每組10只。制備CIA模型,以Western blotting法檢測關(guān)節(jié)滑膜細(xì)胞NF-κB/P65、p-ⅠκBα表達(dá)的情況。結(jié)果:rhTNFR∶Fc治療組和醋酸潑尼松對照組可下調(diào)CIA大鼠滑膜細(xì)胞NF-κB/P65、p-ⅠκBα的表達(dá),與模型組比較均有顯著性差異(P0.05)。結(jié)論:rhTNFR∶Fc可通過降低CIA大鼠關(guān)節(jié)滑膜細(xì)胞NF-κB/P65、p-ⅠκBα蛋白表達(dá)以達(dá)到緩解炎癥的作用。
[Abstract]:Objective: to investigate the effect of recombinant human tumor necrosis factor receptor-antibody fusion protein rhTNFR: Fc on collagen-induced arthritis (CIA). Effects of NF- 魏 B signal expression in synovial cells of model rats. Methods: forty 6-week-old female Wistar rats were randomly divided into blank group according to the random number table method. Model group, prednisone acetate control group and rhTNFR:Fc treatment group, 10 rats in each group. CIA model was made. Western blotting assay was used to detect NF- 魏 B / p65 in synovial cells.
