【摘要】:以聚苯乙烯納米球?yàn)檠芯繉ο?研究基質(zhì)硬度對二維(2D)和三維(3D)培養(yǎng)的乳腺癌細(xì)胞攝取行為的影響。通過光催化聚合反應(yīng)法制備硬度可調(diào)的聚丙烯酰胺凝膠,并在凝膠表面共價(jià)修飾膠原以支持細(xì)胞黏附,構(gòu)建2D細(xì)胞培養(yǎng)模型;選擇Ⅰ型鼠尾膠原構(gòu)建3D細(xì)胞培養(yǎng)模型;分別接種人乳腺癌細(xì)胞MCF-7于聚丙烯酰胺凝膠表面或混懸于膠原凝膠中進(jìn)行3D細(xì)胞培養(yǎng);20 nm和50 nm聚苯乙烯納米球懸液與2D或3D培養(yǎng)的細(xì)胞共孵育,采用激光共聚焦顯微鏡對細(xì)胞形態(tài)和納米球攝取效率進(jìn)行定性和定量考察。本研究發(fā)現(xiàn)在2D和3D培養(yǎng)條件下,基質(zhì)硬度均參與調(diào)控腫瘤細(xì)胞肌動(dòng)蛋白骨架的重構(gòu)并間接影響不同粒徑大小的納米制劑的攝取行為。
[Abstract]:The polystyrene nanospheres were used as the research object.
