靶向納米造影劑C225-USPIO標(biāo)記裸鼠鼻咽癌移植瘤MR成像研究

【摘要】:目的:制備針對(duì)EGFR的靶向納米造影劑C225-USPIO,并檢測(cè)其標(biāo)記裸鼠鼻咽癌移植瘤MR成像(MRI)能力。方法:將西妥昔單抗(C225)與USPIO結(jié)合成C225-USPIO;檢測(cè)造影劑粒徑;選擇EGFR高表達(dá)的鼻咽癌SUNE1 5-8F細(xì)胞株,接種于12只裸鼠的大腿腹側(cè)皮下,待瘤直徑增長(zhǎng)至5mm時(shí),隨機(jī)分實(shí)驗(yàn)組和對(duì)照組各6只,將C225-USPIO或USPIO通過(guò)鼠尾靜脈注射,于注射后0、8、24、72 h經(jīng)磁共振成像儀行T2WI掃描;檢測(cè)腫瘤組織內(nèi)造影劑分布。結(jié)果:C225-USPIO粒徑約45~50 nm;MRI示實(shí)驗(yàn)組8 h T2值較0 h略降低(P0.05),24 h顯著降低(P0.05),72 h無(wú)明顯改變(P0.05)。對(duì)照組8~72 h各時(shí)間點(diǎn)T2值與0 h相比無(wú)明顯改變(P0.05);注射造影劑72 h后腫瘤組織未見(jiàn)明顯鐵顆粒。結(jié)論:C225-USPIO顆粒可通過(guò)毛細(xì)血管,并能運(yùn)用于體內(nèi)磁共振分子成像,可以降低裸鼠移植瘤磁共振T2信號(hào)強(qiáng)度,具有一定的特異性和靶向性。
