成年大鼠視皮層17區(qū)突觸可塑性的探討

【摘要】： 目的： 研究單眼形覺剝奪后反縫對成年期弱視大鼠視皮層17區(qū)突觸可塑性的影響。探討成年期視皮層突觸可塑性的機制。 方法： 將Wistar大鼠分為正常組、弱視組和反縫組。分別飼養(yǎng)至相應時間點時取左側大腦視皮層17區(qū)。應用免疫組織化學法檢測突觸后GABAAR a 1和GluR2亞基的表達水平。透射電子顯微鏡對視皮層17區(qū)第Ⅳ層連續(xù)攝取3,000倍照片,局部定位放大后進行突觸計數(shù)。隨機攝取30,000倍照片,使用Image Proplus 6.0分析軟件對突觸間隙寬度、突觸后致密物厚度、突觸界面曲率進行測量。所有實驗數(shù)據(jù)采用SPSS13.0統(tǒng)計軟件包進行統(tǒng)計學處理。 結果： (1)免疫組織化學結果：P11w時,弱視組和反縫組大鼠視皮層17區(qū)GABAARal的表達較正常組明顯減少(P＜0.001),弱視組較反縫組也明顯減少(P＜0.001)。P13w時,弱視組較反縫組及正常組明顯減少(P＜0.001),而反縫組與正常組比較無明顯差別(P＝0.496),兩個時間點弱視組GluR2的表達較反縫組和正常組均明顯增強(P0.001),反縫組較正常組均明顯較少(P＜0.001)。 (2)突觸超微形態(tài)結構變化：P11w與P13w時,3組大鼠視皮層突觸數(shù)量的差異均有統(tǒng)計學意義,弱視組突觸數(shù)量較反縫組和正常組明顯減少,反縫組較正常組也減少(P＜0.01)。弱視組與反縫組及正常組比較,P11w和P13w時都是突觸間隙明顯增寬,突觸后致密物明顯增厚,突觸界面曲率明顯變小(P＜0.01)。反縫組與正常組比較,P11w時,突觸間隙增寬,突觸后致密物增厚,突觸界面曲率變小(P0.05)；而P13w時,差異無統(tǒng)計學意義(P0.05)。 結論： 視覺系統(tǒng)的可塑性不僅存在于視覺發(fā)育的關鍵期以內(nèi),在關鍵期之后仍然存在。反縫干預能從結構和功能兩方面來使成年大鼠視皮層的可塑性得以改善和恢復。
[Abstract]:Aim: to study the effect of reverse suture after monocular form deprivation on synaptic plasticity in area 17 of visual cortex in adult amblyopia rats. To explore the mechanism of synaptic plasticity in adult visual cortex. Methods: Wistar rats were divided into normal group, amblyopia group and reverse suture group. 17 areas of the left visual cortex were taken at the corresponding time point. The expression of postsynaptic GABAAR a 1 and GluR2 subunits was detected by immunohistochemistry. The fourth layer of visual cortex was taken 3000 times continuously by transmission electron microscope, and the synaptic count was carried out after local localization and amplification. The width of synaptic space, the thickness of postsynaptic density and the curvature of synaptic interface were measured by Image Proplus 6.0 software. All the experimental data were processed statistically by SPSS13.0 software package. Results: (1) Immunohistochemical results: the expression of GABAARal in visual cortex of amblyopia group and reverse suture group was significantly lower than that of normal group at P11 week (P < 0.001).
