β1整合素與聲門型喉癌的關(guān)系

【摘要】: 【中文摘要】目的:檢測β1整合素(intigrinβ1)在聲門型喉癌和正常聲帶組織中的表達并進行定量分析,探討β1整合素在聲門型喉癌發(fā)生、發(fā)展中的作用,以期進一步明確聲門型喉癌的發(fā)病機制,為臨床防治聲門型喉癌提供理論依據(jù)。材料與方法:選取經(jīng)病理診斷確診,且排除其他聲帶病變的聲門型喉癌患者標本30例為實驗組,再按分化程度分為兩組,分別為高分化鱗癌組21例,中、低分化鱗癌組9例;按是否伴頸部淋巴結(jié)轉(zhuǎn)移分為兩組,分別為頸部淋巴結(jié)轉(zhuǎn)移組8例,無頸部淋巴結(jié)轉(zhuǎn)移組22例。取正常聲帶組織標本10例為對照組。所有標本經(jīng)HE染色后,進行MaxVisionTM免疫組織化學(xué)染色,檢測各組中β1整合素的表達情況,并行計算機圖像定量分析。結(jié)果:1.在正常聲帶組織中,β1整合素的表達較弱,而在聲門型喉癌組織中腫瘤細胞的表達則較強。聲門型喉癌組β1整合素的陽性表達率(80.0%)高于正常聲帶組(60.0%),經(jīng)行×列表樣本構(gòu)成比x2檢驗分析,x2=6.494,差異有統(tǒng)計學(xué)意義(P0.05)。2.聲門型喉癌組β1整合素的陽性單位均值(33.9288±8.77570)高于正常聲帶組(23.8556±5.17346),差異有統(tǒng)計學(xué)意義(P0.05)。3.中、低分化鱗癌組β1整合素的陽性單位均值(34.9612±6.95076)雖高于高分化組(32.5840±9.49454),但差異無統(tǒng)計學(xué)意義(P0.05)。4.頸部淋巴結(jié)轉(zhuǎn)移組β1整合素的陽性單位均值(41.1137±5.67882)高于無頸部淋巴結(jié)轉(zhuǎn)移組(30.3287±7.86905),差異有統(tǒng)計學(xué)意義(P0.05)。結(jié)論:1.β1整合素對腫瘤細胞具有很強的特異性和敏感性,可以廣泛用于腫瘤的研究。2.β1整合素在聲門型喉癌組織中存在著較高的表達,與正常聲帶組織相比有明顯差異,且差異有統(tǒng)計學(xué)意義。說明β1整合素表達水平增高與聲門型喉癌的發(fā)生有關(guān),β1整合素的表達與聲門型喉癌呈正相關(guān)。3.β1整合素在中、低分化鱗癌組中的表達雖高于高分化鱗癌組,但差異無統(tǒng)計學(xué)意義,可能與樣本量較小有關(guān)。β1整合素在頸部淋巴結(jié)轉(zhuǎn)移組中的表達明顯高于無頸部淋巴結(jié)轉(zhuǎn)移組,且差異有統(tǒng)計學(xué)意義。表明β1整合素與聲門型喉癌的淋巴結(jié)轉(zhuǎn)移密切相關(guān),提示β1整合素的表達在聲門型喉癌的發(fā)生發(fā)展中起重要作用,促進了腫瘤的侵襲轉(zhuǎn)移。β1整合素的表達增強可能預(yù)示著聲門型喉癌的高侵襲傾向,即具有高黏附性、浸潤生長、易發(fā)生轉(zhuǎn)移等惡性特征,通過對β1整合素的檢測,有助于評估聲門型喉癌的發(fā)生、發(fā)展過程及轉(zhuǎn)移傾向,可作為患者預(yù)后評估的重要指標。
[Abstract]:Objective: to detect the expression and quantitative analysis of 尾 1 integrin (intigrin 尾 1) in glottic carcinoma and normal vocal cord tissues, and to explore the role of 尾 1 integrin in the genesis and development of glottic carcinoma. In order to further clarify the pathogenesis of glottic carcinoma, to provide a theoretical basis for the clinical prevention and treatment of glottic carcinoma. Materials and methods: 30 cases of glottic laryngeal carcinoma diagnosed by pathology and excluding other vocal cord lesions were selected as experimental group and divided into two groups according to the degree of differentiation: 21 cases of well differentiated squamous cell carcinoma group, 9 cases of middle and low differentiated squamous cell carcinoma group. Patients with cervical lymph node metastasis were divided into two groups: cervical lymph node metastasis group (n = 8) and non-cervical lymph node metastasis group (n = 22). 10 normal vocal cord tissues were taken as control group. All samples were stained with HE, MaxVisionTM immunohistochemical staining was performed to detect the expression of 尾 1 integrin in each group, and the quantitative analysis of image was performed. Results: 1. The expression of 尾 1 integrin was weak in normal vocal cord tissues, but strongly expressed in glottic laryngeal carcinoma tissues. The positive expression rate of 尾 1 integrin in glottic carcinoma group (80.0%) was higher than that in normal vocal cord group (60.0%). The X-list sample composition ratio x2 test showed that the positive expression rate of 尾 1 integrin was significantly higher in glottic carcinoma group (80.0%) than that in normal vocal cord group (60.0%) (P0.05). The mean positive unit of 尾 1 integrin in glottic carcinoma group (33.9288 鹵8.77570) was higher than that in normal vocal cord group (23.8556 鹵5.17346), the difference was statistically significant (P0.05). The mean positive unit of 尾 1 integrin in poorly differentiated squamous cell carcinoma group (34.9612 鹵6.95076) was higher than that in well differentiated group (32.5840 鹵9.49454), but there was no significant difference (P0.05). The mean value of 尾 1 integrin positive unit in cervical lymph node metastasis group (41.1137 鹵5.67882) was higher than that in non cervical lymph node metastasis group (30.3287 鹵7.86905) (P0.05). Conclusion: 1. 尾 1 integrin has strong specificity and sensitivity to tumor cells and can be widely used in tumor studies. 2. 尾 1 integrin is highly expressed in glottic laryngeal carcinoma tissues, and 尾 1 integrin is highly expressed in glottic laryngeal carcinoma, and 尾 1 integrin is highly expressed in glottic laryngeal carcinoma. Compared with normal vocal cord tissue, there was significant difference between the two groups. The expression of 尾 1 integrin was positively correlated with glottic carcinoma. 3. The expression of 尾 1 integrin in moderate and poorly differentiated squamous cell carcinoma group was higher than that in well differentiated squamous cell carcinoma group, and the expression of 尾 1 integrin was positively correlated with glottic laryngeal carcinoma, while the expression of 尾 1 integrin in poorly differentiated squamous cell carcinoma group was higher than that in well differentiated squamous cell carcinoma group. The expression of 尾 1 integrin in cervical lymph node metastasis group was significantly higher than that in non cervical lymph node metastasis group, and the difference was statistically significant. The results suggest that 尾 1 integrin is closely related to lymph node metastasis in glottic carcinoma, suggesting that the expression of 尾 1 integrin plays an important role in the genesis and development of glottic carcinoma. The enhanced expression of 尾 1 integrin may predict the high invasion tendency of glottic carcinoma, that is, high adhesion, invasion and growth, easy metastasis and other malignant features, through the detection of 尾 1 integrin, the expression of 尾 1 integrin may indicate the high invasion tendency of glottic carcinoma, and the expression of 尾 1 integrin may indicate the high invasion tendency of glottic carcinoma. It is helpful to evaluate the occurrence, development process and metastasis tendency of glottic carcinoma, and can be used as an important index for prognosis evaluation of patients with glottic carcinoma.
