兔眼脈絡(luò)膜撕脫傷后bFGF與PDGF-A的表達(dá)變化

【摘要】:目的:建立兔眼脈絡(luò)膜撕脫傷模型,觀察不同時(shí)間點(diǎn)堿性成纖維細(xì)胞生長(zhǎng)因子(basic fibroblast growth factor,bFGF)與血小板源性生長(zhǎng)因子(platelet derived growth factor-A,PDGF-A)在脈絡(luò)膜、視網(wǎng)膜及玻璃體內(nèi)的表達(dá)變化,探討脈絡(luò)膜撕脫傷后bFGF與PDGF-A在脈絡(luò)膜增殖修復(fù)中作用,為臨床治療提供新的理論依據(jù)。 方法:40只成年白兔隨機(jī)分成5組,包括正常組,傷后3d、7d、14d、28d實(shí)驗(yàn)組,每組各8只,奇偶數(shù)法確定實(shí)驗(yàn)眼;建立兔眼脈絡(luò)膜撕脫傷模型,于傷后不同時(shí)間點(diǎn)觀察眼底變化并行眼科B超檢查;HE染色光鏡下觀察傷口處脈絡(luò)膜、視網(wǎng)膜形態(tài)學(xué)變化;免疫組化SP法和酶聯(lián)免疫吸附實(shí)驗(yàn)(Enzyme-Linked Immunosorbnent Assay,ELISA)方法分別檢測(cè)脈絡(luò)膜、視網(wǎng)膜及玻璃體內(nèi)bFGF與PDGF-A蛋白表達(dá)量的變化。 結(jié)果:傷后脈絡(luò)膜、視網(wǎng)膜及玻璃體內(nèi)bFGF與PDGF-A表達(dá)上調(diào)。bFGF與PDGF-A蛋白在脈絡(luò)膜、視網(wǎng)膜內(nèi)表達(dá)的平均光密度值及在玻璃體內(nèi)表達(dá)的平均OD值均出現(xiàn)先升高又下降的變化,于傷后3d明顯升高,7d達(dá)到高峰,14d略有回落,仍維持較高水平,28d明顯下降,但高于正常。正常組、各實(shí)驗(yàn)組比較,差異有統(tǒng)計(jì)學(xué)意義(P0.05);傷后7d與14d比較,差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05),傷后7d分別與傷后3d、28d比較,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。 結(jié)論:①bFGF與PDGF-A參與脈絡(luò)膜撕脫傷后脈絡(luò)膜的增殖修復(fù)的發(fā)生發(fā)展,是影響脈絡(luò)膜增殖的因素之一。②脈絡(luò)膜撕脫傷后bFGF與PDGF-A在脈絡(luò)膜、視網(wǎng)膜及玻璃體內(nèi)表達(dá)高峰均出現(xiàn)在7~14天,此時(shí)期處于傷后細(xì)胞增殖期。③本課題為臨床中脈絡(luò)膜撕脫傷后玻璃體視網(wǎng)膜手術(shù)時(shí)機(jī)的選擇、治療干預(yù)提供參考價(jià)值。
[Abstract]:Objective: to establish a rabbit model of choroidal avulsion and observe the expression of basic fibroblast growth factor (basic fibroblast growth factor,bFGF) and platelet-derived growth factor (platelet derived growth factor-A,PDGF-A) in choroid at different time points. To explore the role of bFGF and PDGF-A in the repair of choroid proliferation after choroidal avulsion injury, and to provide a new theoretical basis for clinical treatment. Methods: 40 adult white rabbits were randomly divided into 5 groups, including normal group, 3 days, 7 days, 14 days, 28 days after injury, 8 rabbits in each group, and the experimental eyes were determined by odd and even method. A rabbit model of choroidal avulsion was established, fundus changes were observed at different time points after injury and ophthalmic B-ultrasound examination was performed, choroidal and retinal morphological changes were observed by HE staining under light microscope, and retinal morphological changes were observed at different time points after injury. Immunohistochemical SP method and enzyme-linked immunosorbent assay (Enzyme-Linked Immunosorbnent Assay,ELISA) were used to detect the expression of bFGF and PDGF-A protein in choroid, retina and vitreous. Results: the expression of bFGF and PDGF-A was up-regulated in choroid, retina and vitreous after injury, and the expression of bFGF and PDGF-A protein was up-regulated in choroid. The mean optical density of retina and the OD value of vitreous expression increased first and then decreased at the 3rd day after injury, reached the peak at the 7th day, decreased slightly at the 14th day, still maintained the high level, and decreased significantly on the 28th day after the injury, and the optical density of the retina and the OD value expressed in the vitreous were increased at first and then decreased after the injury. But higher than normal. Normal group, each experimental group, the difference was statistically significant (P0.05); 7 days and 14 days after injury, the difference was not statistically significant (P0.05), 7 days after injury and 3 days, 28 days after injury, the difference was statistically significant (P0.05). Conclusion: 1bFGF and PDGF-A are involved in the occurrence and development of choroidal proliferation and repair after choroidal avulsion injury, which is one of the factors affecting choroidal proliferation. BFGF and PDGF-A are involved in choroidal repair after choroidal avulsion injury, and bFGF and PDGF-A are involved in choroidal repair after choroidal avulsion injury. The peak of expression in retina and vitreous appeared at 7 days and 14 days, which was in the phase of cell proliferation after injury. 3 this study provided reference value for the choice of time for vitreoretinal surgery and treatment intervention after choroidal avulsion injury in clinic.
