中耳加壓治療豚鼠膜迷路積水及其機(jī)制的實(shí)驗(yàn)研究

【摘要】:目的 采用激素法制備豚鼠膜迷路積水動(dòng)物模型[1],將其放入壓力艙內(nèi)進(jìn)行中耳加壓,以探討中耳加壓是否可以抑制膜迷路積水,并通過中耳加壓治療后豚鼠耳蝸水通道蛋白2(aquaporin-2,AQP-2)表達(dá)的變化,進(jìn)一步探討中耳加壓對膜迷路積水影響的機(jī)制。 方法 將30只豚鼠隨機(jī)分為3組,每組10只:對照組不做處理;積水組和加壓組腹腔注射醋酸去氨加壓素1周(4μg/kg/d),制備膜迷路積水模型。且加壓組第2,3周放進(jìn)壓力艙行加壓治療,各組豚鼠生存期都是3周。觀察3組動(dòng)物耳蝸形態(tài)和功能的變化;用免疫組化方法檢測三組豚鼠耳蝸AQP-2的表達(dá)。 結(jié)果 (1)積水組聽腦干反應(yīng)(ABR)聽閾值為[(58.34±7.79) dB SPL]高于加壓組[(33.29±3.26) dB SPL]和對照組[(20.84±2.13)dB SPL](P0.05);(2)膜迷路積水程度:積水組R值為(0.49±0.11)較加壓組(0.36±0.06)膜迷路積水為重P0.05);對照組無膜迷路積水。(3)免疫組化實(shí)驗(yàn):AQP-2主要表達(dá)在蝸管外側(cè)壁、Corti’s器、螺旋神經(jīng)節(jié)細(xì)胞等處,積水組蝸管外側(cè)壁平均灰度值為(134.63±6.64)較對照組(159.36±2.63)、加壓組(147.06±4.91)豚鼠AQP-2的表達(dá)增加(P0.05)。 結(jié)論 1中耳加壓可顯著減輕由醋酸去氨加壓素而引起的豚鼠膜迷路積水并改善了耳蝸功能。 2中耳加壓治療使膜迷路積水豚鼠耳蝸的AQP-2的表達(dá)下降,說明中耳加壓使膜迷路積水減輕的治療機(jī)制可能與AQP-2的表達(dá)下降有關(guān)。
[Abstract]:Objective to establish an animal model of membranous labyrinth hydrops in guinea pigs by hormone method, and put it into the pressure chamber for middle ear pressure, so as to explore whether middle ear pressure can inhibit membranous labyrinth hydrops. The changes of aquaporin-2 (aquaporin-2,AQP-2) expression in guinea pig cochlea after middle ear pressure therapy were used to investigate the mechanism of the effect of middle ear pressure on membranous labyrinthine hydrops. Methods Thirty guinea pigs were randomly divided into 3 groups: control group (n = 10), hydrops group (n = 10), hydrolysate group (n = 10) and hydrops group (n = 10), and hydrops model of membranous labyrinth was established by intraperitoneal injection of desaminopressin acetate (4 渭 g/kg/d). In the second week, the guinea pigs in the compression group were put into the pressure chamber for compression therapy, and the survival time of each group was 3 weeks. The changes of cochlea morphology and function in three groups were observed and the expression of AQP-2 in cochlea of guinea pigs was detected by immunohistochemical method. Results (1) the (ABR) threshold of auditory brainstem response in hydronephrosis group [(58.34 鹵7.79) dB SPL)] was higher than that in pressure group [(33.29 鹵3.26) dB SPL)] and control group (20.84 鹵2.13) dB SPL) (P0.05). (2) the degree of membranous labyrinth hydrops: the R value of hydronephrosis group (0.49 鹵0.11) was higher than that of pressure group (0.36 鹵0.06) (P0.05). (3) Immunohistochemical study: AQP-2 was mainly expressed in the lateral wall of cochlear duct, Corti's organ, spiral ganglion cells and so on. The mean gray value of the lateral wall of the cochlear canal in the hydrops group was (134.63 鹵6.64) higher than that in the control group (159.36 鹵2.63), and the expression of AQP-2 in the pressurized group (147.06 鹵4.91) was increased (P0.05). Conclusion 1 the middle ear pressure can significantly reduce the hydrocephalus caused by deaminopressin acetate and improve the cochlea function in guinea pigs. 2the expression of AQP-2 in cochlea of guinea pigs with membranous hydrolabyrinth decreased after middle ear pressure treatment, indicating that the mechanism of reducing membranous hydrolabyrinth by middle ear pressure may be related to the decrease of AQP-2 expression.
