外周血NF-κB和TNF-α表達(dá)與甲狀腺相關(guān)性眼病相關(guān)性研究

【摘要】:目的:甲狀腺相關(guān)性眼病(Thyroid Associated Ophthalmopathy TAO)是一種與甲狀腺疾病相關(guān)的器官特異性自身免疫性疾病,其發(fā)病機(jī)制尚不明確。目前研究已證明,多種細(xì)胞因子(Cytokines,CK)如干擾素(Interferon,IFN)、白細(xì)胞介素-1(Interleukin -1,IL-1)、IL-4、腫瘤壞死因子(Tumor necrosis factor,TNF)等參與TAO的發(fā)生與發(fā)展。核因子-κB(NCR Clear factor kappa B,NF-κB)是一具有多功能轉(zhuǎn)錄與調(diào)節(jié)作用的蛋白質(zhì)因子,它能與多種細(xì)胞因子和粘附因子基因啟動(dòng)子部位發(fā)生結(jié)合,增強(qiáng)這些基因的轉(zhuǎn)錄和表達(dá)。本課題通過(guò)檢測(cè)NF-κB以及TNF-α在TAO患者血液中的表達(dá),分析其與TAO的相關(guān)性,為臨床治療提供實(shí)驗(yàn)基礎(chǔ)。 方法:研究對(duì)象共160例,分為三組:TAO組54例、Graves。℅raves disease,GD)組52例和正常對(duì)照(CON)組54例。用酶聯(lián)免疫吸附法(ELISA)檢測(cè)血清中TNF-α的水平,蛋白印跡法(western blot)檢測(cè)外周血中有核細(xì)胞中NF-κB的水平。 結(jié)果:①TAO組、GD組和CON組外周血血清中TNF-α濃度分別為6.157±0.653ng/ml、14.088±2.598ng/ml和2.398±0.326ng/ml;與正常對(duì)照組比較,TAO和GD組血清TNF-α濃度顯著增高(P0.01;P0.01);與GD組比較,TAO組血清TNF-α濃度顯著降低(P0.01)。 ②TAO組、GD組和CON組外周血細(xì)胞核蛋白NF-κB的含量分別為0.334±0.490ng/ml、1.186±0.159ng/ml和0.077±0.0005ng/ml;與正常對(duì)照組比較,TAO和GD組外周血NF-κB表達(dá)水平增高(P0.01;P0.01);與GD組比較,TAO組外周血NF-κB表達(dá)水平顯著降低(P0.01)。 結(jié)論:①TAO和GD患者外周血中TNF-α和NF-κB的表達(dá)水平顯著增高。②TAO患者外周血中TNF-α和NF-κB的高表達(dá)沒有特異性。
[Abstract]:Objective: thyroid associated ophthalmopathy (Thyroid Associated Ophthalmopathy TAO) is an organ-specific autoimmune disease associated with thyroid disease. At present, many cytokines (Cytokines,CK), such as interferon (Interferon,IFN), interleukin-1 (Interleukin -1), IL-4, tumor necrosis factor (Tumor necrosis factor,TNF), have been proved to be involved in the pathogenesis and development of TAO. Nuclear factor- 魏 B (NCR Clear factor kappa (NF- 魏 B) is a multifunctional transcription and regulatory protein factor, which can bind to the promoter of many cytokines and adhesion factors, and enhance the transcription and expression of these genes. In this study, we detected the expression of NF- 魏 B and TNF- 偽 in the blood of TAO patients, analyzed their correlation with TAO, and provided the experimental basis for clinical treatment. Methods: one hundred and sixty patients were divided into three groups: TAO group (n = 54), Graves's disease (Graves disease,GD) group (n = 52) and normal control group (CON) group) (n = 54). Enzyme linked immunosorbent assay (ELISA) was used to detect the level of TNF- 偽 in serum and Western blot (western blot) was used to detect the level of NF- 魏 B in nucleated cells. Results: the serum TNF- 偽 concentrations in 1TAO group, GD group and CON group were 6.157 鹵0.653 ng / ml, 14.088 鹵2.598ng/ml and 2.398 鹵0.326 ng / ml, respectively. Compared with the normal control group, the serum TNF- 偽 concentration in TAO and GD groups was significantly higher (P0.01, P0.01), and that in TAO group was significantly lower than that in GD group (P0.01). The contents of nuclear protein NF- 魏 B in 2TAO group, GD group and CON group were 0.334 鹵0.490 ng / ml 1.186 鹵0.159ng/ml and 0.077 鹵0.0005 ng / ml, respectively. Compared with normal control group, the expression of NF- 魏 B in peripheral blood of TAO and GD groups was higher than that in GD group (P0.01), and the expression of NF- 魏 B in TAO group was significantly lower than that in GD group (P0.01). Conclusion: the expression of TNF- 偽 and NF- 魏 B in peripheral blood of patients with 1TAO and GD is significantly higher than that in patients with 2TAO, but the high expression of TNF- 偽 and NF- 魏 B in peripheral blood of 2TAO patients has no specificity.
