【摘要】：目的:研究基質(zhì)金屬蛋白酶-2(MMP-2)在鼻息肉與鼻息肉內(nèi)鏡手術(shù)后囊泡組織中的表達(dá)。 材料與方法:取10例鼻息肉、10例鼻息肉內(nèi)鏡術(shù)后囊泡組織以及10例單純鼻中隔偏曲患者部分下鼻甲(對(duì)照組)。采用免疫熒光法觀察MMP-2蛋白的定位及分布,采用實(shí)時(shí)定量PCR法檢測(cè)MMP-2在三組組織中mRNA的表達(dá),采用免疫印跡蛋白技術(shù)檢測(cè)MMP-2在三組組織中蛋白的定量表達(dá)。 結(jié)果:1.息肉組與囊泡組上皮細(xì)胞、炎性細(xì)胞與腺上皮細(xì)胞MMP-2蛋白高表達(dá),對(duì)照組部分腺上皮細(xì)胞MMP-2蛋白高表達(dá);2.MMP-2在息肉組和囊泡組中的mRNA水平均比對(duì)照組增高(p0.05),MMP-2在息肉組與囊泡組中的mRNA水平無明顯差異; 3.MMP-2在鼻息肉組和囊泡組中的蛋白水平均比對(duì)照組增高(p0.05),MMP-2在鼻息肉組和囊泡組中的蛋白水平無明顯差異。 結(jié)論:MMP-2在鼻息肉與囊泡組織中的表達(dá)上調(diào);術(shù)后抑制囊泡組織過度增生對(duì)預(yù)防鼻息肉復(fù)發(fā)有一定意義;研究MMP-2抑制物對(duì)鼻息肉的治療有一定意義。
[Abstract]:Aim: to investigate the expression of matrix metalloproteinase-2 (MMP-2) in the vesicles of nasal polyps and nasal polyps after endoscopic surgery. Materials and methods: 10 cases of nasal polyps, 10 cases of nasal polyps after endoscopic surgery, and 10 cases of simple nasal septum deviation of partial inferior turbinate (control group). The localization and distribution of MMP-2 protein were observed by immunofluorescence, the expression of mRNA in three groups was detected by real-time quantitative PCR, and the protein expression of MMP-2 in three groups was detected by Western blot technique. The result is 1: 1. The expression of MMP-2 protein in the epithelial cells of polyp group and vesicle group, inflammatory cells and glandular epithelial cells was high, while that of MMP-2 protein in some glandular epithelial cells of the control group was higher than that in the control group. The mRNA level of 2.MMP-2 in polyp group and vesicle group was higher than that in control group (p0. 05). There was no significant difference in mRNA level between polyp group and vesicle group. The protein level of 3.MMP-2 in nasal polyp group and vesicle group was higher than that in control group (p0.05), but there was no significant difference in MMP-2 protein level in nasal polyp group and vesicle group. Conclusion: the expression of MMP-2 is up-regulated in nasal polyps and vesicles, the inhibition of excessive proliferation of vesicular tissue is of significance in preventing the recurrence of nasal polyps, and the study of MMP-2 inhibitors in the treatment of nasal polyps is of some significance.
