【摘要】： 目的：檢測(cè)茄病鐮刀菌感染大鼠角膜早期表面活性蛋白A和D (surfactant protein A and D,SPA、SP-D)的表達(dá),初步探討SP-A、SP-D在真菌性角膜炎固有免疫中的作用。 方法：Wistar大鼠80只隨機(jī)分成4組：空白對(duì)照組、單純損傷組、真菌性角膜炎一組、真菌性角膜炎二組。右眼為實(shí)驗(yàn)眼。空白對(duì)照組5只大鼠,不做任何處理；單純損傷組25只模擬模型的制作不接種茄病鐮刀菌；真菌性角膜炎一組、真菌性角膜炎二組各25只建立茄病鐮刀菌性角膜炎動(dòng)物模型。單純損傷組、真菌性角膜炎一組、真菌性角膜炎二組分別于建模后6h、12h、24h、48h、96h處死5只大鼠。各組分別在裂隙燈顯微鏡下觀察大鼠的眼部變化；病理組織切片觀察角膜組織炎癥程度；(空白對(duì)照組、單純損傷組、真菌性角膜炎一組取右眼角膜,真菌性角膜炎二組取左眼角膜)逆轉(zhuǎn)錄聚合酶鏈?zhǔn)椒磻?yīng)(RT-PCR)檢測(cè)角膜組織SP-A、SP-D mRNA表達(dá)；免疫組化技術(shù)檢測(cè)角膜組織中SP-A、SP-D的蛋白表達(dá)以及共表達(dá)強(qiáng)度。 結(jié)果：(1)裂隙燈檢查：真菌感染角膜6h與單純損傷組比較大鼠眼部沒(méi)有明顯差異,瞼球結(jié)膜充血(+),角膜水腫不明顯；12h瞼球結(jié)膜充血(++),角膜出現(xiàn)水腫；24h瞼球結(jié)膜充血(+++),角膜毛玻璃樣水腫；48h、96h角膜水腫明顯,出現(xiàn)黃白色潰瘍,潰瘍面積大小不等,免疫環(huán)形成。(2)病理組織學(xué)檢查：真菌感染角膜6h與單純損傷組比較沒(méi)有明顯差異,角膜中心上皮層缺如,膠原纖維排列整齊,未見(jiàn)明顯中性粒細(xì)胞浸潤(rùn)；12h基質(zhì)淺層少量中性粒細(xì)胞侵入；24h角膜水腫,角膜中心上皮和前彈力層缺失,基質(zhì)層膠原纖維排列紊亂,基質(zhì)淺層中性粒細(xì)胞侵潤(rùn)增加；48h、96h角膜水腫明顯,角膜基質(zhì)排列更加紊亂,基質(zhì)全層大量中性粒細(xì)胞浸潤(rùn)。(3)免疫組化檢測(cè)：正常大鼠、單純損傷組以及真菌性角膜炎二組角膜上皮細(xì)胞有微弱的SP-A蛋白、SP-D蛋白的表達(dá),三組比較沒(méi)有明顯差異(p0.05)；真菌感染角膜6h角膜上皮細(xì)胞SP-A蛋白及SP-D蛋白表達(dá)沒(méi)有明顯增強(qiáng),與正常角膜、單純損傷組以及真菌性角膜炎二組比較沒(méi)有明顯差異(p0.05),12h后表達(dá)逐漸增強(qiáng),48h最強(qiáng),96h下降,各時(shí)間點(diǎn)總體比較表達(dá)水平差異有顯著性(p0.01),與空白對(duì)照組以及單純損傷組和真菌性角膜炎二組相同時(shí)間點(diǎn)比較表達(dá)水平差異有顯著性(p0.01)。(4) RT-PCR檢測(cè)：正常大鼠、單純損傷組、真菌性角膜炎二組角膜有少量SP-A mRNA、SP-D mRNA的表達(dá),三組比較沒(méi)有明顯差異(p0.05)；真菌感染角膜后SP-A mRNA、SP-D mRNA的表達(dá)量逐漸增加,24h最高,48h、96h下降,各時(shí)間點(diǎn)總體比較表達(dá)水平差異有顯著性(p0.01),與空白對(duì)照組以及單純損傷組和真菌性角膜炎二組相同時(shí)間點(diǎn)比較具有顯著差異(p0.01) 結(jié)論：茄病鐮刀菌感染角膜的早期SP-A、SP-D在角膜組織表達(dá)顯著增加,隨著病情進(jìn)展表達(dá)有所下降,但仍高于單純損傷組。提示SP-A、SP-D可能為茄病鐮刀菌的模式識(shí)別受體,在角膜抵抗茄病鐮刀菌感染早期固有免疫應(yīng)答中發(fā)揮著一定作用。
[Abstract]:Objective: To detect the expression of surfactant protein A and D (SPA, SP-D) in the cornea of rats infected with Fusarium solanacearum at early stage, and to explore the role of SP-A and SP-D in the innate immunity of fungal keratitis.
Methods: 80 Wistar rats were randomly divided into 4 groups: blank control group, simple injury group, fungal keratitis group, fungal keratitis group, right eye, blank control group, 5 rats without any treatment, simple injury group, 25 model rats without inoculation of Fusarium solani, fungal keratitis group, fungal cornea group. Five rats were sacrificed at 6 h, 12 h, 24 h, 48 h and 96 h after the models were established in the simple injury group, fungal keratitis group and fungal keratitis group. (blank control group, simple injury group, fungal keratitis group take right eye cornea, fungal keratitis group take left eye cornea) reverse transcription polymerase chain reaction (RT-PCR) detection of corneal tissue SP-A, SP-D mRNA expression; immunohistochemistry detection of corneal tissue SP-A, SP-D protein expression and co-expression intensity.
Results: (1) Slit lamp examination: There was no significant difference between the fungal infected cornea and the injured cornea at 6 h, but the conjunctiva was hyperemic (+) and the corneal edema was not obvious; the conjunctiva was hyperemic (++) at 12 h, the cornea was edematous; the conjunctiva was hyperemic (++++) at 24 h, and the cornea was ground glass edema at 48 h and 96 h; the corneal edema was obvious and the ulcer was yellow and white. (2) Histopathological examination: there was no significant difference between the fungal infected cornea and the simple injured cornea at 6 h. The central epithelial layer of cornea was absent, the collagen fibers were arranged neatly, and no obvious neutrophil infiltration was found; a small amount of neutrophil invasion was found in the superficial layer of stroma at 12 h; corneal edema at 24 h, and the central cornea was damaged. The loss of the skin and the anterior elastic layer, the disorder of collagen fibers in the stroma, the increase of neutrophil invasion in the superficial layer of the stroma, the obvious corneal edema at 48h and 96h, the disorder of corneal stroma arrangement, and the infiltration of a large number of neutrophils in the whole stroma. (3) Immunohistochemical detection: normal rats, simple injury group and fungal keratitis two groups of corneal epithelial cells. There was no significant difference in the expression of SP-A and SP-D proteins among the three groups (p0.05). The expression of SP-A and SP-D proteins in corneal epithelial cells of fungal infection cornea was not significantly increased at 6 hours, but there was no significant difference compared with normal cornea, simple injury cornea and fungal keratitis (p0.05). The expression gradually increased after 12 hours, the strongest at 48 hours, 96 hours. The expression level of SP-A mRNA and SP-D-mR in cornea of normal rats, simple injury group and fungal keratitis group were significantly different (p0.01). There was no significant difference in the expression of NA among the three groups (p0.05). The expression of SP-A mRNA and SP-D mRNA increased gradually after fungal infection, the highest at 24 hours, the lowest at 48 hours and the lowest at 96 hours. Differences (P0.01)
CONCLUSION: The expression of SP-A and SP-D in the cornea of Fusarium solani infection in early stage increased significantly, but decreased with the progression of the disease, but still higher than that in the simple injury group.
