Ruboxistaurin對(duì)小鼠氧誘導(dǎo)視網(wǎng)膜病變中新生血管抑制作用的形態(tài)學(xué)觀察

【摘要】: 目的從形態(tài)學(xué)上探討不同劑量Ruboxistaurin對(duì)氧誘導(dǎo)視網(wǎng)膜病變新生血管動(dòng)物模型小鼠視網(wǎng)膜新生血管的作用 方法將出生7天的C57BL/6J小鼠隨機(jī)分成:正常對(duì)照組,高氧模型組,藥物處理組和空白對(duì)照組,除正常對(duì)照組之外,其余組放入氧分壓為(75±2)%的氧箱內(nèi)飼養(yǎng)5天后取出,然后藥物治療組小鼠經(jīng)胃灌注不同劑量Ruboxistaurin,空白對(duì)照組灌注相同劑量甲磺酸鹽,于鼠齡17天時(shí)將所有小鼠全部處死,取其眼球做標(biāo)本固定,分別用計(jì)數(shù)突破視網(wǎng)膜內(nèi)界膜的內(nèi)皮細(xì)胞核數(shù)和偶氮藍(lán)(Evans藍(lán))灌注血管造影法比較小鼠視網(wǎng)膜新生血管的情況。結(jié)果氧誘導(dǎo)視網(wǎng)膜病變模型鼠的視網(wǎng)膜中央及遠(yuǎn)周見無血管灌注區(qū),周邊部有大量新生血管形成;治療組與高氧對(duì)照組和空白對(duì)照組相比,突破視網(wǎng)膜內(nèi)界膜血管內(nèi)皮細(xì)胞核明顯減少,差別有顯著意義(P0.01),視網(wǎng)膜新生血管和無灌注區(qū)較高氧組也明顯好轉(zhuǎn)。 結(jié)論氧誘導(dǎo)視網(wǎng)膜病變新生血管動(dòng)物模型建立成功;氧誘導(dǎo)視網(wǎng)膜病變模型鼠視網(wǎng)膜新生血管和無灌注區(qū)較正常組增多;Ruboxistaurin能改善氧誘導(dǎo)視網(wǎng)膜病變小鼠視網(wǎng)膜新生血管的情況。
[Abstract]:Objective to investigate morphologically the effects of different doses of Ruboxistaurin on retinal neovascularization in mice with oxygen-induced retinopathy. Methods C57BL/6J mice were randomly divided into two groups: normal control group. In the hyperoxia model group, the drug treatment group and the blank control group, in addition to the normal control group, the rest groups were fed in oxygen box with oxygen partial pressure of (75 鹵2)% for 5 days. Then the mice in the drug treatment group were perfused with different doses of Ruboxistaurin through the stomach and the control group were infused with the same dose of methanesulfonate. All the mice were killed at the age of 17 days and their eyeballs were fixed. The retinal neovascularization in mice was compared by counting the number of endothelial nuclei and Evans blue perfusion angiography. Results in the oxygen-induced retinopathy model rats, there were no vascular perfusion areas in the central and distal retina and a large number of neovascularization in the peripheral part of the retina, and the treatment group was compared with the hyperoxia control group and the blank control group. The number of endothelial nuclei in the retinal membranes was decreased significantly (P0.01). The retinal neovascularization and non-perfusion areas were also significantly improved compared with the hyperoxia group. Conclusion the animal model of oxygen-induced retinopathy neovascularization was established successfully, and the number of retinal neovascularization and non-perfusion area in oxygen-induced retinopathy model rats was more than that in normal group. Ruboxistaurin can improve the retinal neovascularization in mice with oxygen-induced retinopathy.
