【摘要】：10只健康新西蘭白兔隨機分為實驗組(n=8)和陰性對照組(n=2)。實驗組兔左眼為實驗眼,右眼為空白對照眼。實驗組采用氫化可的松點眼3 d后行角膜基質(zhì)內(nèi)注入赫氏棘阿米巴滋養(yǎng)體和包囊懸液(1×106/ml)0.2 ml。對照組兔角膜基質(zhì)內(nèi)注射等量生理鹽水。于注射后每天記錄兔眼角膜病變情況,刮取角膜組織進行病原學檢測。于感染后不同時間采血和摘取角膜進行抗體效價檢測和病理學檢查。經(jīng)角膜刮片鏡檢和角膜病理切片HE染色觀察證實,實驗眼感染棘阿米巴,并出現(xiàn)棘阿米巴角膜炎典型表現(xiàn)和病理改變。血清抗體效價隨感染時間的延長而升高,于感染后第28天達峰值(A450值為2.2358),而后逐漸下降,并維持穩(wěn)定,直到處死仍高于陰性對照組。陰性對照組血清抗體效價無明顯變化。
[Abstract]:Ten healthy New Zealand white rabbits were randomly divided into two groups: experimental group (n = 8) and negative control group (n = 2). In the experimental group, the left eye was experimental eye and the right eye was blank control eye. In the experimental group, hydrocortisone was used for 3 days and the corneal stroma was injected into the trophozoites of Echinoamoeba Hersoni and the capsule suspension (1 脳 106/ml) 0.2 ml. The control group was injected with the same amount of saline into the cornea stroma. The pathological changes of rabbit cornea were recorded every day after injection and the corneal tissue was scraped for etiological examination. Blood and cornea were collected at different time after infection for antibody titer detection and pathological examination. The experimental eyes were infected with Acanthamoeba and had typical manifestations and pathological changes of Acanthamoeba keratitis. The titer of serum antibody increased with the prolongation of infection time and reached the peak value (A450 = 2.2358) on the 28th day after infection, and then decreased gradually, and remained stable until the death was still higher than that of the negative control group. The titer of serum antibody in negative control group did not change significantly.
