【摘要】：目的 觀察αB-晶狀體蛋白對(duì)大鼠急性高眼壓后視網(wǎng)膜神經(jīng)節(jié)細(xì)胞存活及凋亡蛋白Caspase-3表達(dá)的影響。 方法 3月齡雄性Sprague-Dawley(SD)大鼠,體重250±20g ,160只完全隨機(jī)設(shè)計(jì)分成以下4組:A組(急性高眼壓組),B組(急性高眼壓+玻璃體穿刺組),C組(急性高眼壓+玻璃體注射生理鹽水組),D組(急性高眼壓+玻璃體注射αB-晶狀體蛋白組),應(yīng)用熒光金逆行標(biāo)記計(jì)數(shù)法、免疫熒光法觀察急性高眼壓后各組大鼠視網(wǎng)膜神經(jīng)節(jié)細(xì)胞在不同時(shí)間點(diǎn)的存活率及凋亡蛋白Caspase-3表達(dá)情況。 結(jié)果 急性高眼壓后實(shí)驗(yàn)動(dòng)物右眼(實(shí)驗(yàn)眼)視網(wǎng)膜神經(jīng)節(jié)細(xì)胞開(kāi)始減少,低倍熒光顯微鏡下可見(jiàn)全視網(wǎng)膜節(jié)細(xì)胞均在減少,第一周減少最為明顯,兩周后減少放緩。與其它3組相比,D組在急性高眼壓后1周、2周、3周、4周視網(wǎng)膜神經(jīng)節(jié)細(xì)胞存活率明顯提高,差異具有統(tǒng)計(jì)學(xué)意義(P0.01),其它3組之間,差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05);急性高眼壓后實(shí)驗(yàn)動(dòng)物右眼(實(shí)驗(yàn)眼)Caspase-3免疫熒光染色出現(xiàn)陽(yáng)性表達(dá),與其它3組相比,D組在急性高眼壓后24小時(shí)、48小時(shí)、72小時(shí)、1周的Caspase-3表達(dá)明顯減少,差異具有統(tǒng)計(jì)學(xué)意義(P0.01),其它3組之間,差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。 結(jié)論 αB-晶狀體蛋白對(duì)大鼠急性高眼壓后視網(wǎng)膜神經(jīng)節(jié)細(xì)胞存在保護(hù)作用,能增加視網(wǎng)膜神經(jīng)節(jié)細(xì)胞的存活率。 αB-晶狀體蛋白能抑制急性高眼壓后視網(wǎng)膜神經(jīng)節(jié)細(xì)胞內(nèi)凋亡蛋白Caspase-3的表達(dá),減少細(xì)胞凋亡。
[Abstract]:Objective to observe the effect of 偽 B crystallin on the survival of retinal ganglion cells and the expression of apoptotic protein Caspase-3 after acute intraocular hypertension in rats. Methods 3-month-old male Sprague-Dawley (SD) rats were used. 160 rats weighing 250 鹵20 g were randomly divided into 4 groups: group A (acute intraocular pressure group) and group B (acute intraocular pressure vitreous puncture group) and group C (acute intraocular pressure vitreous injection normal saline group) and D group (acute high intraocular pressure group). The glass body was injected with 偽 B-crystallin group, and the fluorescent gold retrograde labeling method was used. The survival rate of retinal ganglion cells (RGCs) and the expression of apoptotic protein (Caspase-3) in retinal ganglion cells (RGCs) after acute intraocular pressure (IOP) were observed by immunofluorescence method at different time points. Results after acute high intraocular pressure, the retinal ganglion cells in the right eye (experimental eyes) of experimental animals began to decrease, and the total retinal ganglion cells decreased under low power fluorescence microscope. The decrease was most obvious in the first week and slowed down two weeks later. Compared with the other three groups, the survival rate of retinal ganglion cells in group D was significantly higher than that in group D at 1 week, 2 weeks and 3 weeks and 4 weeks after acute intraocular pressure (P0.01), and the survival rate of retinal ganglion cells in group D was significantly higher than that in group D (P0.01). There was no significant difference (P0.05) in the expression of Caspase-3 in the right eye (experimental eyes) of experimental animals after acute intraocular hypertension, and the expression of Caspase-3 in group D was significantly lower than that in group D (24 hours, 48 hours, 72 hours and 1 week after acute high intraocular pressure) compared with the other three groups. The difference was statistically significant (P0.01), the other three groups, the difference was not statistically significant (P0.05). Conclusion 偽 B-crystallin has protective effect on retinal ganglion cells after acute intraocular hypertension in rats. 偽 B-crystallin could inhibit the expression of apoptotic protein Caspase-3 in retinal ganglion cells after acute intraocular hypertension and decrease apoptosis.
