【摘要】： 目的：檢測(cè)SHIP2蛋白在喉鱗狀細(xì)胞癌(喉鱗癌)組織及其細(xì)胞系中的表達(dá),并分析其表達(dá)與喉鱗癌臨床病理特征及預(yù)后之間的關(guān)系。 方法：1).運(yùn)用Western blot技術(shù)檢測(cè)SHIP2蛋白在喉鱗癌細(xì)胞株Hep-2的表達(dá)情況；2).采用免疫組織化學(xué)技術(shù)在54例喉鱗癌及16例癌旁粘膜石蠟組織標(biāo)本中檢測(cè)SHIP2蛋白的表達(dá)水平,并統(tǒng)計(jì)分析SHIP2表達(dá)水平與喉鱗癌臨床病理特征及預(yù)后的關(guān)系。 結(jié)果：SHIP2蛋白在喉鱗癌細(xì)胞株Hep-2中的表達(dá)較上呼吸道永生化粘膜上皮細(xì)胞株NP-69顯著上調(diào)(0.88±0.09VS0.17±0.07)；在喉鱗癌組織中的表達(dá)較癌旁粘膜組織的陽(yáng)性率亦顯著增高(77.78%VS18.75%)；且其表達(dá)水平與喉鱗癌的T分期(T1+T2/T3+T4) (x2=10.119,P=0.006)、臨床分期(Ⅰ期+Ⅱ期/Ⅲ期+Ⅳ期)(x2=13.460,P=0.001)、轉(zhuǎn)移(x2=12.959,P=0.002)及復(fù)發(fā)(x2=11.241,P=0.004)密切相關(guān)；SHIP2高表達(dá)患者較低表達(dá)患者復(fù)發(fā)快(中位復(fù)發(fā)時(shí)間29.346月±3.605月VS51.321月±2.975月,x2=16.336,P0.001),中位生存時(shí)間(41.231月±3.164月)亦顯著短于SHIP2低表達(dá)患者(56.036月±2.267月)(x2=19.893,P0.001)；且SHIP2表達(dá)水平結(jié)合臨床分期、淋巴結(jié)有無(wú)轉(zhuǎn)移對(duì)預(yù)后具有更好的預(yù)測(cè)作用。多因素Cox比例風(fēng)險(xiǎn)回歸模型結(jié)果顯示,SHIP2表達(dá)水平與淋巴結(jié)有無(wú)轉(zhuǎn)移同為喉鱗癌預(yù)后的獨(dú)立影響因素。 結(jié)論：本研究發(fā)現(xiàn)SHIP2蛋白在喉鱗癌組織及細(xì)胞中的表達(dá)顯著升高,并與喉鱗癌的復(fù)發(fā)、轉(zhuǎn)移及預(yù)后密切相關(guān)。這些結(jié)果均提示SHIP2可能在喉鱗癌的發(fā)生、發(fā)展中發(fā)揮了重要作用,有望成為預(yù)測(cè)喉鱗癌復(fù)發(fā)、轉(zhuǎn)移及預(yù)后的重要分子標(biāo)志物及潛在的治療靶點(diǎn)。
[Abstract]:Aim: to detect the expression of SHIP2 protein in laryngeal squamous cell carcinoma (LSCC) tissues and its cell lines, and to analyze the relationship between SHIP2 expression and clinicopathological features and prognosis of laryngeal squamous cell carcinoma (LSCC). Method 1). The expression of SHIP2 protein in laryngeal squamous cell carcinoma cell line Hep-2 was detected by Western blot. The expression of SHIP2 protein was detected by immunohistochemistry in 54 cases of laryngeal squamous cell carcinoma and 16 cases of paraffin tissue of adjacent mucosa.
