S100A7在喉鱗癌組織中表達(dá)的研究

【摘要】:背景與目的 腫瘤的發(fā)生發(fā)展是多基因參與、多因素影響的復(fù)雜過程。在頭頸部惡性腫瘤發(fā)生過程中,上皮細(xì)胞的異常增殖被認(rèn)為與腫瘤的形成緊密相關(guān)。喉鱗癌是最常見的頭頸部惡性腫瘤之一。近年來喉鱗癌的發(fā)病率有逐年增加的趨勢,,雖然在過去的數(shù)年里,經(jīng)綜合治療(手術(shù)+放療+化療)喉癌患者的生存率有所提高,但晚期患者的預(yù)后仍然很差。故早期診斷對病人接受更好的治療、延長生存時間具有重要的意義。迄今為止,尚未發(fā)現(xiàn)確定的生物學(xué)標(biāo)志物提示潛在的腫瘤形成的惡化過程,同位素標(biāo)記相對量和絕對量及液相色譜分析法顯示S100A7可作為頭頸部鱗狀細(xì)胞癌的標(biāo)志性蛋白。既往亦有研究顯示S100A7在乳腺癌、肺鱗癌、食管鱗癌等多種鱗癌中過表達(dá),S100A7蛋白在細(xì)胞增殖、信號傳導(dǎo)、炎性反應(yīng)及血管形成等病理生理過程中起著重要的作用,而關(guān)于S100A7在喉鱗癌組織中的表達(dá)情況及S100A7能否作為輔助診斷及判斷預(yù)后的一個標(biāo)志尚未被證實。本研究通過檢測S100A7在喉鱗癌組織中的表達(dá)情況,初步探討其與喉鱗癌發(fā)生發(fā)展的關(guān)系。 材料與方法 標(biāo)本來源:所有喉鱗癌組織、喉鱗癌轉(zhuǎn)移灶組織、正常喉粘膜組織均采自于重慶醫(yī)科大學(xué)附屬第二醫(yī)院耳鼻咽喉頭頸外科住院部住院患者。喉鱗癌及其轉(zhuǎn)移灶診斷依據(jù)病理診斷證實。收集的標(biāo)本置于-80℃冰箱保存待用。 方法:1.采用RT-PCR方法和Western-blot方法分別檢測喉鱗癌組織、喉鱗癌轉(zhuǎn)移灶組織、正常喉粘膜組織及不同病理分化程度的喉鱗癌組織中S100A7基因及蛋白表達(dá)情況;2.采用統(tǒng)計軟件SPSS19.0對數(shù)據(jù)進(jìn)行統(tǒng)計分析,以a=0.05為檢驗標(biāo)準(zhǔn),P0.05表示在統(tǒng)計學(xué)上該差異具有統(tǒng)計學(xué)意義。 結(jié)果 1.經(jīng)RT-PCR檢測到喉鱗癌組織、喉鱗癌轉(zhuǎn)移灶組織、正常喉粘膜組織中均有S100A7基因的表達(dá),且在喉鱗癌組織中高表達(dá),差異具有統(tǒng)計學(xué)意義(P0.05);S100A7基因轉(zhuǎn)錄水平在不同病理分化程度的標(biāo)本中不一樣,其表達(dá)量在高分化、中分化、低分化喉鱗癌組織中依次遞增,差異具有統(tǒng)計學(xué)意義(P0.05)。 2.Western-blot檢測到喉鱗癌組織、喉鱗癌轉(zhuǎn)移灶組織、正常喉粘膜組織中均有S100A7蛋白的表達(dá),且在喉鱗癌組織中高表達(dá),差異具有統(tǒng)計學(xué)意義(P0.05);S100A7蛋白在高分化、中分化、低分化喉鱗癌組織中的表達(dá)量依次遞增,差異具有統(tǒng)計學(xué)意義(P0.05)。 結(jié)論 1. S100A7在喉鱗癌中高表達(dá),在喉鱗癌原發(fā)灶及轉(zhuǎn)移灶中表達(dá)量高于正常喉粘膜組織。S100A7可能與喉鱗癌的發(fā)生、轉(zhuǎn)移有關(guān)。 2.喉鱗癌組織中,S100A7的表達(dá)量與病理分化程度有關(guān),S100A7可能參與了喉鱗癌的分化進(jìn)展過程。
[Abstract]:Background and objective tumorigenesis and development is a complex process in which multiple genes are involved and many factors are involved. The abnormal proliferation of epithelial cells is thought to be closely related to the formation of malignant tumors in head and neck. Laryngeal squamous cell carcinoma (LSCC) is one of the most common malignant tumors in head and neck. In recent years, the incidence of laryngeal squamous cell carcinoma (LSCC) has been increasing year by year. Although the survival rate of patients with LSCC has increased in the past few years, the prognosis of advanced patients is still very poor. Therefore, early diagnosis is of great significance for patients to receive better treatment and prolong their survival time. Up to now, no identified biomarkers have been found to indicate the potential progression of tumorigenesis. The relative and absolute amounts of isotopic markers and liquid chromatography showed that S100A7 could be used as the iconic protein of squamous cell carcinoma of the head and neck. Previous studies have shown that the overexpression of S100A7 protein plays an important role in cell proliferation, signal transduction, inflammatory response and angiogenesis in breast cancer, lung squamous cell carcinoma, esophageal squamous cell carcinoma and other squamous cell carcinomas. The expression of S100A7 in laryngeal squamous cell carcinoma (LSCC) and whether S100A7 can be used as a marker for diagnosis and prognosis have not been confirmed. In this study, the expression of S100A7 in laryngeal squamous cell carcinoma (LSCC) was detected to explore the relationship between S100A7 expression and laryngeal squamous cell carcinoma (LSCC). Materials and methods: all laryngeal squamous cell carcinoma tissues, metastatic laryngeal squamous cell carcinoma tissues and normal laryngeal mucosa were collected from inpatients in the department of otolaryngology and head and neck surgery of the second affiliated Hospital of Chongqing Medical University. The diagnosis of laryngeal squamous cell carcinoma and its metastasis was confirmed by pathological diagnosis. The collected specimens were stored in-80 鈩
