【摘要】：目的:研究慶大霉素對(duì)C57BL/6J小鼠聽(tīng)力及耳蝸內(nèi)毛細(xì)胞帶狀突觸CaV1.3鈣通道蛋白數(shù)量的影響,探討聽(tīng)力損失與其劑量相關(guān)性變化之間的關(guān)系及意義。方法:采用固定劑量[100mg/(kg·d)]的氨基苷類抗生素(慶大霉素)對(duì)C57BL/6J小鼠每日進(jìn)行腹腔注射造模,分別在注射后的第7天、第14天、第28天(注射14d停藥14d)應(yīng)用ABR分別測(cè)得小鼠的聽(tīng)力。免疫熒光方法觀察耳蝸基膜內(nèi)毛細(xì)胞帶狀突觸CaV1.3鈣通道蛋白的分布和表達(dá)(其中0天為空白對(duì)照組),內(nèi)毛細(xì)胞突觸前后膜分別采用CtbP2和CaV1.3進(jìn)行免疫熒光標(biāo)記。結(jié)果:隨著注射藥物天數(shù)的增長(zhǎng)ABR反應(yīng)閾值明顯提高,所有小鼠內(nèi)毛細(xì)胞數(shù)量、形態(tài)均無(wú)明顯變化,但內(nèi)毛細(xì)胞帶狀突觸CaV1.3鈣離子通道蛋白量的表達(dá)存在顯著差異(P0.01)。CaV1.3鈣離子通道蛋白在慶大霉素腹腔注射CaV1.3/CtBP2表達(dá)數(shù)量在第7天時(shí)略有增加,在第14天時(shí)其表達(dá)數(shù)量明顯減少,第28天與第14天比較CaV1.3/CtBP2表達(dá)數(shù)量有所增加,但低于正常。結(jié)論:在氨基苷類抗生素毒性環(huán)境中耳蝸內(nèi)毛細(xì)胞CaV1.3蛋白表現(xiàn)為先增加后減少再增加的趨勢(shì),提示內(nèi)毛細(xì)胞帶狀突觸CaV1.3蛋白數(shù)量的增加可能存在對(duì)這類藥物毒性的代償作用,而隨著藥物毒性作用時(shí)間的增加,這種失代償作用表現(xiàn)為聽(tīng)力下降,可能是損傷聽(tīng)力的機(jī)制之一。
[Abstract]:Aim: to study the effects of gentamicin on the number of calcium channel protein (CaV1.3) in the band synapses of C57BL / 6J mice and to explore the relationship between hearing loss and its dose-dependent changes. Methods: C57BL / 6J mice were intraperitoneally injected with gentamicin at a fixed dose of [100mg/ (kg d)]. The hearing of C57BL / 6J mice was measured on the 7th, 14th and 28th day after injection. The distribution and expression of CaV1.3 calcium channel protein (CaV1.3) in hair cells of cochlear basal membrane were observed by immunofluorescence method. CtbP2 and CaV1.3 were used to label the synaptic membrane of the inner hair cell before and after treatment with CtbP2 and CaV1.3, respectively. Results: with the increase of the days of injection, the ABR reaction threshold increased significantly, and the number and morphology of hair cells in all mice did not change significantly. However, there was a significant difference in the expression of CaV1.3 calcium channel protein in the inner hair cells (P0.01) .CaV1.3 calcium channel protein in gentamicin intraperitoneal injection of CaV1.3 / CtBP2 increased slightly on the 7th day, and decreased significantly on the 14th day. On the 28th day, compared with the 14th day, the expression of CaV1.3 / CtBP2 was increased, but lower than that of the normal. Conclusion: in the toxic environment of aminoside antibiotics, the expression of CaV1.3 protein in cochlear hair cells increased first and then decreased, suggesting that the increase in the number of band synaptic CaV1.3 protein in the inner hair cells may have a compensatory effect on the toxicity of these drugs. With the increase of the time of drug toxicity, the decompensation may be one of the mechanisms of hearing loss.
