去整合素Echistatin對(duì)糖尿病兔后發(fā)性白內(nèi)障模型中晶狀體上皮細(xì)胞間質(zhì)轉(zhuǎn)分化及膠原合成的影響

【摘要】:目的觀察去整合素Echistatin對(duì)糖尿病兔晶狀體摘出術(shù)后后囊膜混濁(posterior capsular opacification,PCO)的影響,探討Echistatin對(duì)晶狀體上皮細(xì)胞(lens epithelial cells,LEC)間質(zhì)轉(zhuǎn)分化及膠原合成的抑制作用。方法應(yīng)用四氧嘧啶耳緣靜脈注射建立糖尿病兔模型,隨機(jī)分組并行晶狀體摘出術(shù),術(shù)畢前房分別注入0.2 mL高壓滅菌蒸餾水(對(duì)照組),7.5 mg·L-1Echistatin(7.5 mg·L-1組)、10.0 mg·L-1Echistatin(10.0 mg·L-1組),每組又分為10 d和6周2個(gè)時(shí)間點(diǎn)(n=4)。裂隙燈顯微鏡觀察各組糖尿病兔術(shù)后10 d和6周PCO分級(jí)情況,同時(shí)摘取術(shù)眼應(yīng)用RT-PCR法檢測(cè)晶狀體后囊膜上α-平滑肌肌動(dòng)蛋白(α-smooth muscle actin,α-SMA)和Ⅳ型膠原mRNA表達(dá)情況。結(jié)果術(shù)后10 d,各組間α-SMA mRNA和IV型膠原mRNA表達(dá)差異均無(wú)統(tǒng)計(jì)學(xué)意義(均為P0.05)。術(shù)后6周,Echistatin干預(yù)組PCO分級(jí)均明顯低于對(duì)照組(P=0.024);7.5 mg·L-1Echistation組α-SMA mRNA表達(dá)為3.97±0.76,對(duì)照組為9.06±3.40,10.0 mg·L-1組為6.80±3.15,7.5 mg·L-1組明顯低于其余兩組(P=0.002、0.024);Ⅳ型膠原mRNA表達(dá)對(duì)照組為30.88±20.13,7.5 mg·L-1組為8.62±6.99,10.0 mg·L-1組為5.76±6.73,Echistatin干預(yù)組Ⅳ型膠原mRNA表達(dá)均低于對(duì)照組(P=0.005、0.002)。結(jié)論去整合素Echistatin對(duì)糖尿病兔晶狀體摘出術(shù)后PCO的發(fā)生和發(fā)展有一定的抑制作用,可能與其抑制晶狀體后囊膜上α-SMA和Ⅳ型膠原的mRNA表達(dá)有關(guān)。
[Abstract]:Objective to observe the effect of Echistatin on (posterior capsular opacification of posterior capsular opacification after lens extraction in diabetic rabbits, and to investigate the inhibitory effect of Echistatin on interstitial transdifferentiation and collagen synthesis of lens epithelial cells (lens epithelial cells). Methods Diabetic rabbit model was established by intravenous injection of alloxan into ear margin. At the end of the operation, 0.2 mL high pressure sterilized distilled water (control group) was injected with 7.5 mg L -1 Echistatin (7.5 mg L -1 group) and 10.0 mg L -1 Echistatin (10.0 mg L -1 group) respectively.
