【摘要】: 目的:探討Netrin-1蛋白在C57BL/6J小鼠氧誘導(dǎo)視網(wǎng)膜病變(OIR)模型中視網(wǎng)膜中的表達(dá),及其與視網(wǎng)膜新生血管形成的相關(guān)性。 方法:隨機(jī)取16只C57BL/6J新生小鼠分為正常組和高氧誘導(dǎo)組,每組8只。通過(guò)高氧誘導(dǎo)的方法使用C57BL/6J小鼠建立氧誘導(dǎo)視網(wǎng)膜病變(OIR)模型。小鼠17日齡時(shí),先于各組隨機(jī)取出2只小鼠熒光造影后行視網(wǎng)膜鋪片,通過(guò)觀察視網(wǎng)膜新生血管的形態(tài)來(lái)鑒定視網(wǎng)膜新生血管模型;剩余小鼠全部處死后取出眼球固定,行冰凍切片,免疫組織化學(xué)技術(shù)檢測(cè)Netrin-1蛋白質(zhì)在視網(wǎng)膜中的表達(dá)。 結(jié)果:(1)熒光造影視網(wǎng)膜鋪片觀察視網(wǎng)膜血管形態(tài):正常組17日齡小鼠視網(wǎng)膜鋪片顯示視網(wǎng)膜血管分布呈均勻的網(wǎng)狀結(jié)構(gòu)。OIR模型組17日齡小鼠視網(wǎng)膜視盤(pán)周邊見(jiàn)大片無(wú)灌注區(qū)。無(wú)灌注區(qū)周?chē)梢?jiàn)新生血管,伴明顯熒光滲漏。(2)免疫熒光結(jié)果;OIR模型小鼠建立成功,17日齡的OIR模型組小鼠視網(wǎng)膜處于新生血管增生期,視網(wǎng)膜淺層可見(jiàn)Netrin-1蛋白明顯表達(dá),而17日齡正常組小鼠視網(wǎng)膜未見(jiàn)Netrin-1蛋白表達(dá)。 結(jié)論:OIR小鼠視網(wǎng)膜新生血管期Netrin-1在視網(wǎng)膜淺層表達(dá),提示Netrin-1可能參與視網(wǎng)膜新生血管的形成過(guò)程。
[Abstract]:Aim: to investigate the expression of Netrin-1 protein in the retina of C57BL/6J mice induced by oxygen induced retinopathy and its correlation with retinal neovascularization. Methods: sixteen C57BL/6J newborn mice were randomly divided into normal group and hyperoxia induced group with 8 mice in each group. The model of oxygen-induced retinopathy was established in C57BL/6J mice by hyperoxia induction. At the age of 17 days, the retinal neovascularization model was identified by observing the morphology of retinal neovascularization, and the rest of the mice were killed and their eyeballs were fixed. The expression of Netrin-1 protein in retina was detected by immunohistochemistry. Results the retinal vascular morphology was observed by fluorescein angiography (FG): the retinal vascular distribution was uniform in the normal group of 17 days old mice. In the oir model group, there was a large area of no perfusion around the optic disc of the 17 day old mouse retina. Neovascularization was seen around the no perfusion area, accompanied by obvious fluorescence leakage. (2) the results of immunofluorescence showed that the OIR model mice of 17 days old were in the phase of neovascularization, and the Netrin-1 protein was obviously expressed in the superficial layer of the retina. However, no expression of Netrin-1 protein was found in the retina of 17-day-old normal mice. Conclusion the expression of Netrin-1 in retinal neovascularization phase of OIR mice in the superficial layer of retina suggests that Netrin-1 may be involved in the formation of retinal neovascularization.
