HSP90和eNOS在激素性青光眼發(fā)病中的作用

【摘要】:目的 探討HSP90和內(nèi)皮型一氧化氮合酶(eNOS)在糖皮質(zhì)激素性青光眼發(fā)病中的作用,以及與視網(wǎng)膜神經(jīng)節(jié)細(xì)胞的凋亡之間的內(nèi)在聯(lián)系。為今后對(duì)糖皮質(zhì)激素性青光眼的進(jìn)一步研究和治療提供理論依據(jù)。 方法 28只新西蘭大白兔,隨機(jī)分成4組,對(duì)照組、實(shí)驗(yàn)A組、B組、C組,每組7只,實(shí)驗(yàn)組隔日定時(shí)左眼球結(jié)膜下注射地塞米松0.5mg(0.1ml)。對(duì)照組隔日左眼球結(jié)膜下注射等量生理鹽水。各組隔日定時(shí)測左眼眼壓。A組用藥兩周后進(jìn)行指標(biāo)檢測,B組和C組繼續(xù)隔日定時(shí)左眼球結(jié)膜下注射地塞米松0.5mg(0.1ml)共四周。四周后B組進(jìn)行各項(xiàng)指標(biāo)測定,C組停藥觀察眼壓變化后進(jìn)行指標(biāo)測定。免疫組化觀察每組小梁網(wǎng)中eNOS的陽性表達(dá)和視網(wǎng)膜中HSP90的陽性表達(dá),TUNEL法檢測視網(wǎng)膜神經(jīng)節(jié)細(xì)胞(RGCs)的凋亡情況。 結(jié)果 正常對(duì)照組的眼壓在各個(gè)時(shí)間段變化差別無統(tǒng)計(jì)學(xué)意義(P0.05)。模型組中用藥4次后眼壓開始升高,隨著用藥時(shí)間的延長眼壓升高明顯。A、B、C組用藥兩周后眼壓升高明顯,與對(duì)照組比較差別有統(tǒng)計(jì)學(xué)意義(P0.05)。實(shí)驗(yàn)組B組和C組用藥四周后眼壓升高到實(shí)驗(yàn)的峰值,與對(duì)照組比較差異性顯著(P0.01)。視網(wǎng)膜中的HSP90陽性表達(dá)和小梁網(wǎng)中的eNOS陽性表達(dá)隨著用藥時(shí)間的延長與對(duì)照組比較陽性表達(dá)明顯減少,A組與對(duì)照組比較差別有統(tǒng)計(jì)學(xué)意義(P0.05),B、C組與對(duì)照組比較差異性顯著(P0.01)。A、B、C組RGCs細(xì)胞隨著用藥時(shí)間的延長眼壓的增高凋亡逐漸增多。 結(jié)論 HSP90和eNOS的陽性表達(dá)都與視網(wǎng)膜神經(jīng)節(jié)細(xì)胞的凋亡之間存在顯著的負(fù)相關(guān)。充分說明兩者的含量減少在糖皮質(zhì)激素性青光眼發(fā)病中起著至關(guān)重要的作用,為今后對(duì)糖皮質(zhì)激素性青光眼的進(jìn)一步研究和治療提供理論依據(jù)。
[Abstract]:Purpose To investigate the role of HSP90 and endothelial nitric oxide synthase (Enos) in the pathogenesis of glucocorticoid glaucoma and the relationship between them and the apoptosis of retinal ganglion cells. To provide theoretical basis for the further study and treatment of glucocorticoid glaucoma. Method Twenty-eight New Zealand white rabbits were randomly divided into 4 groups: control group and experimental group A and B with 7 rabbits in each group. The experimental group was injected with dexamethasone 0.5 mg / L 0.1 ml / L subconjunctival dexamethasone every other day. In the control group, the same amount of saline was injected under the bulbar conjunctiva of the left eye every other day. Left intraocular pressure (IOP) in group A was measured two weeks later. The indexes of group B and group C continued to be measured by subconjunctival injection of dexamethasone (0.5 mg / ml) for four weeks. Four weeks later, the indexes were measured in group B and group C after stopping drug and observing the change of intraocular pressure. The expression of eNOS in trabecular meshwork and the expression of HSP90 in retina were observed by immunohistochemical staining and Tunel method was used to detect the apoptosis of retinal ganglion cells (RGCs). Result There was no significant difference in intraocular pressure (IOP) in normal control group (P 0.05). The IOP in the model group began to increase after 4 times of medication, and the IOP increased significantly with the prolongation of the time of administration. The IOP in group A BX C increased significantly after two weeks of treatment, and there was a significant difference between the two groups (P 0.05). The intraocular pressure in group B and group C increased to the experimental peak after four weeks, and the difference was significant compared with the control group (P 0.01). HSP90 positive expression in retina and eNOS positive expression in trabecular meshwork were significantly decreased compared with control group. There was significant difference between group A and control group (P 0.05). The apoptosis of RGCs cells increased with the prolongation of intraocular pressure (IOP). Conclusion There was a significant negative correlation between the positive expression of HSP90 and eNOS and the apoptosis of retinal ganglion cells. It is fully explained that the decrease of the content of both plays an important role in the pathogenesis of glucocorticoid glaucoma and provides a theoretical basis for the further study and treatment of glucocorticoid glaucoma in the future.
