PLUNC蛋白和TLR-4在鼻息肉組織中的表達(dá)及相關(guān)性研究

【摘要】:【目的】旨在研究PLUNC蛋白和TLR-4在鼻息肉和正常鼻粘膜組織中的表達(dá)情況及存在的差異性,分析PLUNC蛋白和TLR-4在鼻息肉組織中表達(dá)的相關(guān)性,進(jìn)一步探討二者在鼻息肉病因?qū)W及疾病進(jìn)展中的意義,為研究CRSwNP的發(fā)病機(jī)制提供科學(xué)依據(jù),奠定良好的實(shí)驗(yàn)基礎(chǔ),同時(shí)也為CRSwNP的臨床治療提供新思路。 【方法】本實(shí)驗(yàn)標(biāo)本主要收集自2013年03月~2013年11月在山西醫(yī)科大學(xué)附屬汾陽(yáng)醫(yī)院耳鼻咽喉科的住院患者,分鼻息肉組和對(duì)照組兩組,術(shù)中取CRSwNP患者篩竇內(nèi)息肉組織標(biāo)本45例作為鼻息肉組,鼻中隔偏曲患者正常中鼻甲粘膜組織標(biāo)本30例作為對(duì)照組。實(shí)驗(yàn)主要采用western印跡法,半定量檢測(cè)正常鼻粘膜組織和鼻息肉組織中PLUNC蛋白和TLR-4的表達(dá)情況。所有實(shí)驗(yàn)數(shù)據(jù)均采用SPSS17.0統(tǒng)計(jì)軟件包進(jìn)行統(tǒng)計(jì)學(xué)分析。 【結(jié)果】①PLUNC蛋白與TLR-4在鼻息肉組織與正常鼻粘膜組織中均有表達(dá),PLUNC蛋白在鼻息肉組與正常對(duì)照組中的相對(duì)積分光密度值分別為0.054±0.022、0.535±0.266,TLR-4在兩組中的相對(duì)積分光密度值分別為0.571±0.144、0.080±0.078。②PLUNC蛋白和TLR-4在鼻息肉組與對(duì)照組鼻粘膜組織的表達(dá)均存在差異且差異有統(tǒng)計(jì)學(xué)意義(p0.05),TLR-4在鼻息肉組織中表達(dá)高于對(duì)照組,而PLUNC蛋白在鼻息肉組織中的表達(dá)則低于對(duì)照組;③鼻息肉組織中PLUNC蛋白與TLR-4表達(dá)存在負(fù)相關(guān)性(r=-0.724,p0.05)。 【結(jié)論】①PLUNC蛋白在鼻息肉組織中低表達(dá),而在正常鼻粘膜中高表達(dá),表明了具有抗炎殺菌活性的PLUNC蛋白表達(dá)受限可能是導(dǎo)致鼻息肉發(fā)生及發(fā)展的又一重要因素。②鼻息肉組織中TLR-4表達(dá)高于正常對(duì)照組,提示感染性因素可能在鼻息肉的發(fā)病過(guò)程中起著重要的作用。TLR-4高表達(dá)可能促進(jìn)了鼻息肉的發(fā)生及發(fā)展。③鼻息肉組織中PLUNC蛋白和TLR-4表達(dá)存在負(fù)相關(guān)性,提示TLR-4可能通過(guò)啟動(dòng)相應(yīng)的信號(hào)轉(zhuǎn)導(dǎo)通路,同時(shí)產(chǎn)生炎癥介質(zhì)、細(xì)胞因子,,從而直接抑制PLUNC蛋白的表達(dá),造成PLUNC蛋白的產(chǎn)生及分泌下降。
[Abstract]:[objective] to study the expression and difference of PLUNC protein and TLR-4 in nasal polyps and normal nasal mucosa, and to analyze the correlation between PLUNC protein and TLR-4 expression in nasal polyps. To further explore the significance of the two in the etiology and disease progression of nasal polyps, to provide a scientific basis for the study of the pathogenesis of CRSwNP, to lay a good experimental foundation, and to provide new ideas for the clinical treatment of CRSwNP. [methods] the patients in the Department of Otolaryngology, Fenyang Hospital affiliated to Shanxi Medical University from March 2013 to November 2013 were collected and divided into two groups: nasal polyp group and control group. During the operation, 45 cases of polyps in ethmoid sinus of CRSwNP patients were taken as nasal polyps group, 30 cases of normal middle turbinate mucosa of patients with deviation of nasal septum as control group. The expression of PLUNC protein and TLR-4 in normal nasal mucosa and nasal polyps was detected by western blot. All experimental data were statistically analyzed by SPSS17.0 statistical software package. [results] the relative integral optical density of 1PLUNC and TLR-4 in nasal polyps and normal nasal mucosa were 0.054 鹵0.022, 0.535 鹵0.266, respectively. The expression of TLR-4 protein and TLR-4 in nasal mucosa of nasal polyp group and control group were significantly higher than that in nasal polyp tissue, and the expression of TLR-4 in nasal polyp tissue was significantly higher than that in control group, and the expression of TLR-4 in nasal polyp tissue was significantly higher than that in control group. However, the expression of PLUNC protein in nasal polyps was lower than that in control group (P < 0.05). There was a negative correlation between PLUNC protein and TLR-4 expression in nasal polyps. [conclusion] the expression of 1PLUNC protein is low in nasal polyps, but high in normal nasal mucosa. The results suggested that the limited expression of PLUNC protein with anti-inflammatory bactericidal activity might be another important factor leading to the occurrence and development of nasal polyps. 2 the expression of TLR-4 in nasal polyps was higher than that in normal controls. The results suggest that infectious factors may play an important role in the pathogenesis of nasal polyps. The high expression of TLR-4 may promote the occurrence and development of nasal polyps. 3. There is a negative correlation between PLUNC protein and TLR-4 expression in nasal polyps. It is suggested that TLR-4 may directly inhibit the expression of PLUNC protein and decrease the production and secretion of PLUNC protein by activating the corresponding signal transduction pathway and simultaneously producing inflammatory mediators and cytokines.
