【摘要】：目的:探討核因子-κB(NF-κB)與缺氧誘導(dǎo)因子-1(HIF-1)在阻塞性睡眠呼吸暫停低通氣綜合征(OSAHS)的激活情況。方法 :研究對象分為輕中度OSAHS組(16例)、重度OSAHS組(14例)及對照組(30例),通過實(shí)時熒光定量PCR法(Real-time)及Western blotting法分別測定外周血單個核細(xì)胞(PBMC)的NF-κB、HIF-1m RNA水平和NF-κB、HIF-1蛋白水平。結(jié)果:重度OSAHS組NF-κBp65m RNA及NF-κBp65蛋白表達(dá)、HIF-1αm RNA及HIF-1α蛋白表達(dá)均顯著高于對照組及輕中度OSAHS組(均P0.01);NF-κBp65 m RNA及NF-κBp65蛋白表達(dá)輕中度OSAHS組與對照組無明顯差異(P值分別為0.068及0.254),輕中度OSAHS組HIF-1αm RNA明顯高于對照組(P0.05)但HIF-1α蛋白表達(dá)無明顯差異(P=0.777)。NF-κBp65m RNA及HIF-1αm RNA的表達(dá)與呼吸暫停低通氣指數(shù)(AHI)呈正相關(guān)(r=0.493,P=0.006,r=0.508,P=0.004),與夜間最低血氧飽和度(LSa O2)呈負(fù)相關(guān)(r=-0.488,P=0.006,r=-0.46,P=0.011)。NF-κBp65蛋白表達(dá)與AHI呈正相關(guān)(r=0.669,P0.001)。HIF-1α蛋白表達(dá)與AHI、氧減指數(shù)(ODI)呈正相關(guān)(r=0.645,P=0.001;r=0.480,P=0.018)。NF-κBp65m RNA與HIF-1αm RNA呈正相關(guān)(r=0.543,P=0.002),NF-κBp65、HIF-1α蛋白表達(dá)呈正相關(guān)(r=0.716,P0.001)。結(jié)論:OSAHS患者NF-κB及HIF-1的基因和蛋白表達(dá)均出現(xiàn)上調(diào),且患者病情越重上調(diào)越明顯,表明OSAHS的發(fā)病與兩者均有關(guān)。
[Abstract]:Aim: to investigate the activation of nuclear factor- 魏 B (NF- 魏 B) and hypoxia inducible factor-1 (HIF-1) in obstructive sleep apnea hypopnea syndrome (OSAHS). Methods: the subjects were divided into mild and moderate OSAHS group (n = 16), severe OSAHS group (n = 14) and control group (n = 30). The levels of NF- 魏 BmHIF-1 RNA and NF- 魏 BHIF-1 protein in peripheral blood mononuclear cells (PBMCs) were measured by real-time quantitative PCR and Western blotting. Results: the expression of NF- 魏 Bp65m RNA and NF- 魏 Bp65 protein, HIF-1 偽 m RNA and HIF-1 偽 protein in severe OSAHS group were significantly higher than those in control group and mild to moderate OSAHS group (P 0.01). There was no significant difference in the expression of NF- 魏 Bp65 m RNA and NF- 魏 Bp65 protein between mild and moderate OSAHS group and control group (P = 0.068 and 0.254 respectively). The expression of HIF-1 偽 m RNA in mild to moderate OSAHS group was significantly higher than that in control group (P0.05), but there was no significant difference in the expression of HIF-1 偽 protein. The expression of HIF-1 偽 Bp65m RNA and HIF-1 偽 m RNA were positively correlated with apnea hypopnea index (AHIH) and negatively correlated with nocturnal minimum oxygen saturation (LSAO _ 2), and negatively correlated with nocturnal minimum oxygen saturation (LSAO _ 2). There was a positive correlation between the expression of AHI and the protein expression of AHI. HIF-1 偽 was positively correlated with the expression of HIF-1 偽 RNA and the protein expression of NF-kappa p65 HIF-1 偽 was positively correlated with the protein expression of rr 0.645P0. 080P 0. 018t. NF- 魏 Bp65m RNA was positively correlated with the expression of NF- 魏 Bp65HIF-1 偽 protein, and the expression of HIF-1 偽 was positively correlated with the expression of HIF-1 偽 RNA (r0. 543P0. 002HIF-1 偽). Conclusion the gene and protein expressions of NF- 魏 B and HIF-1 were up-regulated in patients with OSAHS, and the more severe the disease was, the more obvious the expression of NF- 魏 B and HIF-1 were, which indicated that the pathogenesis of OSAHS was related to both of them.
