快速老化小鼠聽(tīng)功能及耳蝸組織神經(jīng)生長(zhǎng)因子受體酪氨酸激酶受體A的增齡性變化

【摘要】:目的:探討快速老化小鼠聽(tīng)覺(jué)功能以及耳蝸組織中神經(jīng)生長(zhǎng)因子受體酪氨酸激酶受體A(nerve growth factor receptor TrkA,NGFR TrkA)表達(dá)的增齡性變化。方法 :選用3、5、7月齡的快速老化小鼠亞系8(SAMP8)作為觀察對(duì)象。應(yīng)用聽(tīng)覺(jué)誘發(fā)反應(yīng)儀檢測(cè)其8 kHz短純音聽(tīng)覺(jué)腦干反應(yīng)閾值;應(yīng)用NGFR TrkA免疫組化染色檢測(cè)其耳蝸組織染色光密度進(jìn)行分析。結(jié)果:SAMP8聽(tīng)性腦干反應(yīng)閾值隨著月齡增加而顯著增高(P0.05);NGFR TrkA蛋白在不同月齡快速老化小鼠耳蝸組織中均有表達(dá),SAMP8耳蝸組織中的NGFR TrkA蛋白染色光密度隨著月齡增加而顯著降低(P0.05)。結(jié)論:NGFR TrkA蛋白的表達(dá)水平隨著快速老化小鼠聽(tīng)覺(jué)功能增齡性減退而降低,說(shuō)明NGFR TrkA可能與維持耳蝸的功能狀態(tài)有關(guān)。
[Abstract]:Aim: to investigate the aging changes of auditory function and expression of nerve growth factor receptor tyrosine kinase receptor (A(nerve growth factor receptor TrkANGFR) in cochlea of rapidly aging mice. Methods: the rapid aging mouse subline 8 (SAMP8), aged from 3 to 5 to 7 months old, was selected as the observation object. Auditory evoked response apparatus was used to detect the auditory brainstem response threshold of 8 kHz short tone auditory brainstem, and NGFR TrkA immunohistochemical staining was used to detect the optical density of the cochlea tissue staining. Results the threshold of auditory brainstem response of SAMP8 was significantly increased with the increase of age. The expression of P0.05 TrkA protein in cochlea of different months of aging mice was significantly increased. The light density of NGFR TrkA protein in the cochlea of SAMP8 cochlea decreased significantly with the increase of age. Conclusion the expression level of TrkA protein decreases with the aging of auditory function in rapidly aging mice, suggesting that NGFR TrkA may be related to maintaining the functional state of cochlea.
